摘要

目的:基于BDNF/TrkB通路介导的自噬探讨胃袖状切除术(SG)对肥胖大鼠子代认知功能的影响.方法:选用清洁级雌性SD大鼠30只,分为对照组、肥胖组、SG组,各组大鼠与普通饲料喂养的健康雄性大鼠交配,待各组交配获得的子代大鼠成长至3周龄,Morris水迷宫实验检测大鼠子代认知功能,Nissl染色、H&E染色检测大鼠子代海马中Nissl小体数量,试剂盒检测各组大鼠子代血清中总胆固醇(TG)、甘油三酯(TC)及游离脂肪酸(FFA)含量,Western blotting检测大鼠子代海马组织中BDNF、TrkB、Beclin1及LC3 Ⅱ蛋白水平,免疫组织化学染色检测海马组织中BDNF、TrkB水平,透射电镜检测自噬小体数量.结果:与对照组相比,肥胖组大鼠子代逃避潜伏期延长,穿越平台的次数减少,海马组织Nissl小体数量减少,血清中TG、TC及FFA含量增加,海马组织中BDNF、TrkB、Beclin1及LC3 Ⅱ蛋白水平减少,自噬小体数量减少;与肥胖组相比,SG组大鼠子代逃避潜伏期缩短,穿越平台次数增加,海马组织Nissl小体数量增加,血清中TG、TC及FFA含量减少,海马组织中BDNF、TrkB、Beclin 1及LC3 Ⅱ蛋白水平增加,自噬小体数量增加.结论:SG可能通过激活BDNF/TrkB通路介导的自噬改善肥胖大鼠子代认知功能.

Abstract

Objective To investigate the effects of sleeve gastrectomy(SG)on cognitive function in offspring of obese rats based on autophagy mediated by BDNF/TrkB pathway.Methods Thirty clean female SD rats were selected and divided into control group,obesity group and SG group.The rats in each group were mated with healthy male rats fed ordinary diet.After the offspring of the mating rats in each group grew up to 3 weeks old.The cognitive function of offspring was detected by Morris water maze experiment.Nissl and H&E staining were used to detect the number of Nissl bodies in the hippocampus of rat off-spring.The contents of total cholesterol(TG),triglyceride(TC)and free fatty acid(FFA)in serum of rat offspring in each group were detected by the kit.The protein levels of BDNF,TrkB,Beclin1 and LC3 Ⅱ in offspring hippocampal tissues were detected by Western blotting,the levels of BDNF and TrkB in hippocampal tissues were detected by immunohistochemical staining,and the number of autophagosomes was detected by transmission electron microscopy.Results Compared with the control group,the progeny escape latency of the obese rats was prolonged and the frequency of crossing the platform was reduced,the number of Nissl bodies in hippocampus decreased,and the contents of TG,TC and FFA in serum increased,the protein levels of BDNF,TrkB,Beclin1 and LC3 Ⅱ in hippocampus decreased,the number of autophagosomes decreased;Compared with the obese group,the offspring of SG group had shorter escape latency and increased platform crossing times,the number of Nissl bodies in hippocampus increased,and the contents of TG,TC and FFA in serum decreased,the protein levels of BDNF,TrkB,Beclin1 and LC3 Ⅱ in hippocampus were increased,the number of autophagosomes increased.Conclusion SG can improve the cogni-tive function of offspring of obese rats by activating autophagy mediated by BDNF/TrkB pathway.
