目的:研究泛素特异性肽酶1(ubiquitin specific peptidase 1,USP1)在卵巢癌中的表达及其与患者临床病理特征的相关性,探讨USP1对卵巢癌细胞恶性表型的影响.方法:生物信息学分析USP1在卵巢癌患者中的表达水平及与患者临床分期的关系;免疫组化检测USP1在卵巢癌患者和正常卵巢组织中的表达情况,分析其表达水平与患者临床特征之间的关系;通过慢病毒感染的方式构建过表达USP1的SKOV3细胞株,利用CCK-8实验和克隆形成实验检测细胞增殖和克隆形成能力,利用Transwell实验检测细胞迁移和侵袭能力.结果:卵巢癌组织中的USP1表达水平明显高于正常卵巢组织,并与卵巢癌患者病理类型、腹膜转移以及原位肿瘤的大小或侵犯程度相关,与Ki67、ER、PR、P53的表达水平不相关;体外实验结果表明USP1促进卵巢癌细胞增殖、克隆形成、迁移和侵袭能力.结论:USP1在卵巢癌组织中表达增强,并与患者病理类型、腹膜转移以及临床分期相关,USP1促进卵巢癌细胞恶性表型进展.
Abstract
Objective To investigate the expression of ubiquitin-specific peptidase 1(USP1)in ovarian cancer and its cor-relation with clinicopathologic features of patients,as well as to explore the impact of USP1 on the malignant phenotype of ovarian cancer cells.Methods The expression level of USP1 in ovarian cancer patients and its relationship with clinical stage were analyzed by bioinformatics.Immunohistochemistry was used to detect the expression of USP1 in ovarian cancer patients and normal ovarian tissues,and to analyze the relationship between its expression level and the clinical characteristics of the patients.The SKOV3 cell line overexpressing USP1 was constructed by lentiviral infection,and the cell proliferation and clone formation abilities were detected by CCK-8 and clone formation assay,while the migration and invasion abilities were detected by Transwell assay.Results The expression level of USP1 in ovarian cancer tissues was significantly higher than that in normal ovarian tissues,and was correlated with the pathological type,peritoneal metastasis and the size or invasion degree of tumor in situ,but was not correlated with the expression levels of Ki67,ER,PR and P53.The results of in vitro experiments showed that USP1 promoted the proliferation,colony formation,migration and invasion ability of ovarian cancer cells.Conclusion USP1 expression was enhanced in ovarian cancer tissues and correlated with patients'pathological type,peritoneal metastasis,and clinical stage,and USP1 promoted the progression of the malignant phenotype of ovarian cancer cells.
