The Tetraploid Induction and Identification of Primula oreodoxa Franch
Investigations were carried out on the effects of colchicine treatment on differentiation and tetraploid induction of callus of Primula oreodoxa in vitro culture.The results showed colchicine treatment had obvious inhibition effects on differentiation of callus,and increased the death rate of callus;The highest induction rate,23.08%,was obtained by treating callus in the MS medium with colchicine at 30mg· L-1for 10 days.Compared to diploid plants,the autotetraploid plants had larger,thicker and deeper green leaves,and were larger in stomatal size and compacted in appearance.The chromosome numbers of diploid cells was 2n =2x =24.The autotetraploid cells was 2n =4x =48.
