为研究四倍体亚洲百合品种花粉生活力低的原因,本试验以3个四倍体亚洲百合品种布鲁拉诺(Brunello,2n =4x =48)、瓦迪索(Val Di Sole,2n =4x =48)、穿梭(Tresor,2n=4x=48)为试材,采用卡宝品红染色常规压片法对花粉母细胞减数分裂行为进行观察,并对其花粉生活力进行了测定.结果表明,四倍体亚洲百合品种花粉母细胞在减数分裂过程中出现较多异常现象,包括在后期Ⅰ、后期Ⅱ和末期Ⅱ出现滞后染色体,在末期Ⅰ出现染色体桥,在后期Ⅱ出现分裂不同步、不均等分离,在末期Ⅰ、四分体时期出现微核,以及出现了三核四分体等.品种布鲁拉诺花粉母细胞减数分裂异常频率为10.22%、穿梭为21.48%、瓦迪索为33.15%.布鲁拉诺的花粉生活力明显高于其它2个品种,为81.03%,穿梭为66.03%,瓦迪索为48.81%.花粉母细胞减数过程中存在的异常现象可能是导致四倍体亚洲百合品种花粉败育的主要原因之一.研究为百合多倍体育种及无花粉新品种选育奠定一定基础.
Meiosis Observation of Pollen Mother Cells and Determination of Pollen Viability of Tetraploid Asiatic Lily Cultivars
To study the factors of lower pollen viability of tetraploid Asiatic lily cultivars,the meioses of pollen mother cells of three tetraploid Asiatic lily cultivars (2n =4x =48),‘ Brunello’,‘Val Di Sole’ and ‘Tresor’,were observed with the method of ordinary tablet compressing staining with modified fuchsin basic liquid.Their pollen viabilities were also determined.The results indicated that there were some abnormal meiosis phenomena in pollen mother cells of the three tetraploid Asiatic lily cultivars,including lagging chromosomes in anaphase Ⅰ,anaphase Ⅱ and telophase Ⅱ,chromosome bridges in telophase Ⅰ,asynchronous separation and unequal separation in anaphase Ⅱ,micronucleus in telophase Ⅰ and tetrad,and tetrad with three nuclei etc..The abnormal meiosis rates of pollen mother cells of ‘Brunello’,‘Tresor’ and ‘Val Di Sole’ were 10.22%,21.48% and 33.15% respectively.The pollen germination rate of ‘ Brunello’ was 81.03%,significantly higher than those of the other two cultivars,which were 66.03% and 48.81% in ‘ Tresor’ and ‘ Val Di Sole’ respectively.Abnormal meioses of pollen mother cells of tetraploid Asiatic lily cultivars were supposed to be one of the main reasons of the pollen abortion,providing the basis for the breeding of ployploid non-pollen cultivars in lily.
