Study on the Separation of Phellinus Igniarius Protoplast
The effects of different enzymolysis conditions on Phellinus igniarius protoplast yield were studied while using the mycelia as research materials in order to optimize ideal conditions of separating Phellinus igniarius protoplast effectively.The four factors and three levels orthogonal test were used.Results showed that under the condition of 1.5 % lywallzyme solution,strain age 10 d,0.6 mol · L-1 mannitol as osmotic stabilizers,enzymolysis time 3 h,enzymolysis temperature 30 ℃,rotational speed of enzymolysis 100 r · min-1,Phellinus igniarius protoplast yield was 4 625 ten thousands protoplasts · mL-1,it was the highest of all the treatments.The treatment optimized can be used in the separation of Phellinus igniarius protoplast,it will lay the foundation for breeding Phellinus igniarius'high-yielding strains of various bioactive substances and related genetic research.
