Genetic Diversity and Comprehensive Evaluation of the Phenotypic Characteristics of Auricularia cornea Germplasm Resources
To study the variation,distribution,and diversity of phenotypic characteristics in Auricularia cornea germplasm resources,we analyzed the relationship between characteristics,and identified the germplasms with superior evaluation indicators.A total of 56 Chinese A.cornea germplasms were precisely identified for 18 phenotypic characteristics.The Shannon-Weaver diversity index and coefficient of variation were used to analyze the genetic diversity of these phenotypic traits.Comprehensive evaluations of A.cornea germplasms were conducted using correlation analysis,cluster analysis,principal component analysis and stepwise regression.The results showed that the genetic diversity index of A.cornea germplasm resources ranged from 0.090 to 1.978,while the variation in quantitative characteristics spanned from 4.00%to 40.39%.Most phenotypic traits exhibited varying degrees of correlation,with some overlapping trait information.Cluster analysis,conducted at a genetic similarity coefficient of 0.47,categorized the germplasms into five distinct groups.Principal component analysis transformed the 18 phenotypic characteristics into 7 principal components,or comprehensive factors,achieving a cumulative contribution rate of 74.75%.The principal component comprehensive scores(F values)of the 56 germplasms ranged from-3.91 to 2.21,further categorizing the germplasms into 6 groups,some of which overlapped with those identified in the cluster analysis.Based on the F values,5 germplasms with excellent phenotypic characteristics were selected for further exploration and utilization.Combined with the stepwise regression analysis method,8 phenotypic characteristics were identified as the key indicators for comprehensive evaluation of A.cornea:the mycelium(spawn)color change of the fungus stick,the predominant ventral side color and the dorsal side color of fruiting body,the ventral side color and the dorsal side color of the dried fruiting body,the grain of the fruiting body,the maturity period of the fruiting body,and the length of the fruiting body.This study provides an important basis for the efficient exploration and utilization of A.cornea germplasm resources and the breeding of new varieties.
