基于网络药理学探讨升陷汤治疗慢性心力衰竭的作用机制

目的 基于网络药理学分析方法探索升陷汤治疗慢性心力衰竭的作用机制.方法 利用TCMSP、Pubchem、Swiss Target Prediction筛选升陷汤的活性成分和靶点,通过GeneCard、OMIM、Drugbank数据库获取并筛选与慢性心力衰竭相关的作用靶点,利用STRING数据库分析蛋白质相互作用关系,并构建网络,David数据库将靶点基因进行基因本体、京都基因和基因组百科全书生物通路富集分析.结果 共筛选得到56种活性成分,潜在作用靶点141个,获得山柰酚、槲皮素、豆甾醇、异鼠李素、木犀草素、黄芪皂苷类为核心活性成分,PTGS2、STAT3、TP53、EGFR、MMP9为核心靶点,关键通路包括AGE-RAGE信号通路、PI3K-Akt信号通路、IL-17信号通路、HIF-1信号通路、TNF信号通路.结论 升陷汤治疗慢性心力衰竭主要物质基础可能是黄酮类、甾醇类、皂苷类,其潜在作用机制为抑制心肌细胞凋亡、改善心室重塑、抑制炎症反应及氧化应激等方面,未来应开展分子研究进一步验证.
Mechanism of Shengxian Decoction in the Treatment of Chronic Heart Failure Based on Network Pharmacology
Objective To explore the mechanism of Shengxian decoction in the treatment of chronic heart failure based on network pharmacology methods.Methods TCMSP,Pubchem and Swiss Target Prediction were used to screen the active ingredients and targets of Shengxian decoction.GeneCard,OMIM and Drugbank databases were used to obtain and screen the targets associated with chronic heart failure.Protein interaction was analyzed using STRING platform,network was constructed,and gene ontology and Kyoto Encyclopedia of Genes and Genomes biological pathway enrichment analysis were performed on target genes using David database.Results A total of 56 active ingredients were screened and 141 potential targets were identified.The core active ingredients were kaempferol,quercetin,stigmasterol,isorhamnetin,luteolin,and astragalosides,with PTGS2,STAT3,TP53,EGFR and MMP9 as core targets.Key pathways included AGE-RAGE signaling pathway,PI3K-Akt signaling pathway,IL-17 signaling pathway,HIF-1 signaling pathway and TNF signaling pathway.Conclusion The main substance basis for the treatment of chronic heart failure with Shengxian decoction may be flavonoids,sterols and saponins.Its potential mechanism of action that to inhibit myocardial cell apoptosis,improve ventricular remodeling,inhibit inflammatory response,and oxidative stress.In the future,molecular research should be conducted for further validation.
