Expression of PCNA in Uterine Corpus Endometrial Carcinoma and Its Clinical Significance
Objective To investigate the value of proliferating cell nulcear antigen(PCNA)gene in uterine corpus endometrial carcinoma(UCEC)tissue,and to analyze its relationship with clinicopathological features and potential biological functions.Methods The expression data of PCNA mRNA in UCEC and related pathological parameters were obtained from The Cancer Genome Atlas(TCGA)data.A total of 20 UCEC tissue samples and 20 normal endometrial tissue samples cured in the First Affiliated Hospital of Zhengzhou University from January 2020 to December 2022 were selected.Immunohistochemistry was used to detect the expression of PCNA protein in the above tissues.Univariate and multivariate Cox regression were used to analyze the prognostic value of PCNA in UCEC patients.The relationship between PCNA expression and immune cell infiltration was analyzed using TIMER database.The gene set enrichment analysis(GSEA)software was used to predict the related pathways of PCNA gene expression.STRING database was used to investigate the interaction between related proteins.Results The expression levels of PCNA gene mRNA and encoded protein were up-regulated in UCEC tissues(P<0.001),and had diagnostic value.Immunohistochemical staining showed that PCNA protein was highly expressed in the nucleus of UCEC.The results of Cox regression analysis showed that age,clinical stage,primary therapy outcome,histological type,residual tumor,histological grade,tumor invasion,radiation therapy were potential prognostic factors for UCEC(P<0.001).PCNA gene expression levels were correlated with the abundance of immune-infiltrating cells.GSEA analysis showed that the high expression of PCNA was enriched in the cell cycle,homologous recombination,DNA repair and other pathways.Conclusion PCNA gene may be a potential diagnostic and prognostic marker for UCEC.
