子痫前期患者胎盘组织中缺氧诱导因子-1α表达及其与血管生成的关系

目的 分析子痫前期患者胎盘组织中缺氧诱导因子-1α(HIF-1α)表达及其与血管生成的关系。方法 前瞻性选取2019年1月至2023年1月在洛阳市妇幼保健院接受治疗并成功分娩的80例子痫前期患者作为研究对象,其中轻症患者48例(轻症组),重症患者32例(重症组),另外选取50例同期健康孕妇作为对照组。于全部孕妇分娩后取其全层胎盘组织,检测并记录胎盘组织微血管密度,检测HIF-1α mRNA表达水平,计算HIF-1α阳性表达率。测定孕产妇的活化部分凝血活酶时间(APTT)、血浆凝血酶原时间(PT)、纤维蛋白原(Fib)等凝血相关指标水平。分析HIF-1α相对表达量和胎盘组织中血管生成的关系。结果 重症组胎盘组织中微血管密度低于轻症组和对照组,且轻症组低于对照组(P<0。05)。重症组HIF-1α mRNA、Fib水平高于轻症组和对照组,APTT、PT低于轻症组和对照组,轻症组HIF-1αmRNA、Fib水平高于对照组,APTT、PT低于对照组(P<0。05)。重症组HIF-1α总阳性表达率高于轻症组与对照组,轻症组高于对照组。子痫前期患者胎盘组织中HIF-1α相对表达量与血管生成呈负相关(r<0,P<0。05)。结论 HIF-1α在子痫前期患者的胎盘组织中呈异常过表达,且表达水平和胎盘组织血管生成呈负相关。
Expression of Hypoxia Inducible Factor-1α in Placenta of Preeclampsia Patients and its Relationship with Angiogenesis
Objective To analyze the relationship between the expression of hypoxia inducible factor 1α(HIF-1α)and angiogenesis in placenta of patients with preeclampsia.Methods A total of 80 cases of preeclampsia who were treated in Luoyang Maternal and Child Health Hospital from January 2019 to January 2023 and delivered successfully were prospectively selected as the research objects,including 48 cases of mild patients(mild group)and 32 cases of severe patients(severe group),and 50 cases of healthy pregnant women in the same period were selected as the control group.After delivery,all pregnant women were taken from the entire layer of placental tissue,and the microvessel density of placental tissue was measured and recorded.The expression level of HIF-1α mRNA was measured,and the positive expression rate of HIF-1α was calculated.The levels of activated partial thromboplastin time(APTT),plasma Prothrombin time(PT),fibrinogen(Fib)and other coagulation related indicators in pregnant and lying in women were measured.The relationship between the relative expression of HIF-1α and angiogenesis in placental tissue was analyzed.Results The microvascular density in the placental tissue of the severe group was lower than mild group and the control group,and the mild group was lower than the control group(P<0.05).The levels of HIF-1α mRNA and Fib in the severe group were higher than those in the mild group and control group,while APTT and PT were lower than those in the mild group and control group,the levels of HIF-1α mRNA and Fib in the mild group were higher than those in the control group,while APTT and PT were lower than control group(P<0.05).The total positive expression rate of HIF-1 α in the severe group was higher than mild and control groups,and the mild group was higher than control group(P<0.05).The relative expression level of HIF-1 α in placental tissue of preeclampsia patients was negatively correlated with angiogenesis(r<0,P<0.05).Conclusion HIF-1 α is abnormally overexpressed in placental tissue of preeclampsia patients,and the expression level is negative related to placental tissue angiogenesis.
