Effect of Carbachol on the Function of Pulmonary Microvascular Endothelial Cells in Rats with Hemorrhagic Shock
Objective To observe the effect of carbachol on pulmonary microvascular endothelial cells in rats with hemorrhagic shock.Methods Thirty healthy male SD(SPF grade)rats were randomly divided into three groups as follows at 10 rats per group:sham shock group(SS group),haemorrhagic shock group(HS group),haemorrhagic shock group+carbachol group(CBL group).The animal models of haemorrhagic shock were made according to the Wiggers modified method.Six hours after the preparation of the models in each group,blood was collected via the right femoral artery to detect the level of tumour necrosis factor-α(TNF-α),and the lung tissues of the lower lobe of the right lung were taken to detect the levels of Src-suppressed protein kinase C substrate(SSeCKS)and E-selectin proteins,and the lower lobe of the right lung was taken to be examined by transmission electron microscopy.Results The SS group expressed a small amount of TNF-α,SSeCKS and E-selectin,and their expression was elevated in the HS and CBL groups(P<0.05),and their expression was reduced in the CBL group compared with the HS group(P<0.05).The cell membrane surface of lung microvascular endothelial cells in the SS group was smooth,and the morphology and structure of cell membrane,cytoplasm,and nucleus were normal.The lung microvascular endothelial cells in the HS group showed cellular edema,necrosis,and apoptosis manifestations(folding of the cell membrane surface,unevenness of the cell membrane surface,edema of organelles,and deepening of heterochromatin staining in the border set,etc.).In the CBL group,compared with the SS group,there were cellular damages but they were significantly better than that in the HS group.Conclusion Carbachol can exert a protective effect on pulmonary micro vascular endothelial cells of rats in hemorrhagic shock.It may also attenuate damage to vascular endothelial cells by inhibiting inflammatory cell synthesis and release of inflammatory mediators.
