Diagnostic Value of miR-299-5p Combined with CD44 in Children with Nonsyndromic Cleft Lip and Palate
Objective To explore the diagnostic value of miR-299-5p combined with CD44 in children with nonsyndromic cleft lip and palate. Methods Eighty children with non syndromic cleft lip and palate admitted to the Third People's Hospital of Puyang from July 2019 to July 2022 were selected.Among them,40 children with cleft lip or without cleft palate were recorded as group A,and 40 children with simple cleft palate were recorded as group B.Another 40 healthy children during the same period of physical examination in hospital were recorded as group C.The relative expression levels of miR-299-5p and CD44 were measured by real-time fluorescence quantitative RT-PCR and western blot, respectively. The receiver operating characteristic(ROC)of the subjects was drawn, and the diagnostic efficacy of miR-299-5p and CD44 in children with nonsyndromic cleft lip and palate was evaluated according to the area under the curve(AUC).Results The relative expression of miR-299-5 p in group A and group B was lower than that in group C,and the difference was statistically significant(P<0.05).The relative expression of CD44 in group A and group B was lower than that in group C,the differences were statistically significant(P<0.05).ROC analysis showed that the sensitivity of miR-299-5p and CD44 in the diagnosis of nonsyndromic cleft lip and palate in children were 72.50%,62.50%and 82.50%.The specificity were 72.50%,82.50%and 80.00%respectively.AUC were 0.716(95%CI:0.573-0.859),0.718(95%CI:0.572-0.862),0.821(95%CI:0.716-0.965),respectively,and their combined prediction efficiency was higher than that of single index(P<0.05).Conclusion The diagnostic efficacy of miR-299-5p and CD44 in children with non syndromic cleft lip and palate is good.
