Relationship Between LncRNA FER1L4 and Multi-drug Resistance of Gastric Cancer Cells and Its Mechanism
Objective To explore the relationship between long non-coding RNA(lncRNA)FER1L4 and multi-drug resistance of gastric cancer cells and related potential mechanism.Methods SGC7901,SGC7901 doxorubicin resistant cell line(SGC7901/ADR)and SGC7901 vincristine resistant cell line(SGC7901/VCR)were cultured.SGC7901/ADR cells were divided into control A and test A group,and SGC7901/VCR cells were divided into control B and test B group.SGC7901 was used as parent group.Plvx-Puro vector was transfected into parent group,control group A and control group B by lentivirus transfection,while plvx-FER1L4A-puro vector was transfected into test A and B group.MTT method was used to detect the sensitivity of each group cells to ADR and VCR.Real-time fluorescent quantitative PCR was used to detect the expressions of lncRNA FER1L4 and trefoil factor 1(TFF1)mRNA,and the protein expressions of TFF1 and NF-κB p65 were detected by Western blot.Results Compared with the parent group,the 50%inhibitory concentrations(IC50)of ADR in control A and test A group were higher(P<0.05),and that in test A was lower than that in control A group(P<0.05).Compared with the parent group,the IC50 of ADR in control B and test B group were higher(P<0.05),and that in test B was lower than that in control B group(P<0.05).Compared with the parent group,the expressions of FER11L4 and TFF1 in control A,B group were decreased(P<0.05),and in test A,B group were increased(P<0.05),while the expression of p65 protein was increased in control A,B and test A,B group(P<0.05).Compared with control A,B group,the expression of FER1L4 and TFF1 in test A,B group was increased(P<0.05),while the expression of p65 protein was decreased(P<0.05).Conclusion LncRNA FER1L4 can promote the expression of tumor suppressor gene TFF1 and inhibit the multi-drug resistance of gastric cancer cells.
