Expression of H3K9ac,PD-L1 and VISTA in Different Pathological Features of Breast Cancer Patients and the Value of Their Combined Detection in Predicting Poor Prognosis of Breast Cancer
Objective To investigate the expression of histone H3 lysine residue 9 acetylation(H3K9ac),programmed death ligand 1(PD-L1)and V-domain immunoglobulin suppressor of T-cell activation(VISTA)in different pathological features of breast cancer patients and the value of their combined detection in predicting poor prognosis of breast cancer.Methods A total of 130 cases of breast cancer patients admitted to the First People's Hospital of Pingdingshan from May 2018 to July 2020 were selected.The expressions of H3K9ac,PD-L1 and VISTA in breast cancer tissues and adjacent tissues were detected by immunohistochemistry.The expressions of H3K9ac,PD-L1 and VISTA in breast cancer tissues and adjacent tissues were compared.The expressions of H3K9ac,PD-L1 and VISTA in different pathological features of breast cancer were compared.After 3 years of follow-up,the patients were divided into poor prognosis and good prognosis according to metastasis,recurrence and death.The expressions of H3K9ac,PD-L1 and VISTA in breast cancer patients with poor prognosis and good prognosis were compared.The predictive value of combined detection of H3K9ac,PD-L1 and VISTA expression for poor prognosis of breast cancer was analyzed.Results The high expression rates of H3K9ac,H3K4me3,H3K27ac,PD-L1,and VISTA in cancer tissues were higher than those in para-cancerous tissues(P<0.05).The high expression rates of H3K9ac,PD-L1,and VISTA were different among different tissue differentiation levels,T stages,N stages,and the number of positive lymphocytes(P<0.05).After 3 years of follow-up,among 130 patients with breast cancer,7 cases(5.38%)were lost to follow-up,22 cases had poor prognosis and 101 cases had good prognosis.The high expression rates of H3K9ac,PD-L1,and VISTA in patients with poor prognosis were higher than those in patients with good prognosis(P<0.05).The AUC of H3K9ac,PD-L1 and VISTA combined detection for predicting poor prognosis of breast cancer patients was 0.902,which was higher than the above indicators were detected separately 0.714,0.866 and 0.781(P<0.05).Conclusion H3 K9ac,PD-L1 and VISTA are highly expressed in the cancer tissues of breast cancer patients,and their high expressions are related to the degree of differentiation,stage and number of lymphocytes.The combined detection of H3K9ac,PD-L1 and VISTA has a high predictive value for poor prognosis of breast cancer patients.
