Expression of Myeloperoxidase in Peripheral Blood Neutrophils and Its Clinical Significance in Patients with MPO-AAV
Objective To investigate the expression of myeloperoxidase(MPO)in peripheral blood neutrophils and its clinical significance in patients with MPO-anti-neutropil cytoplasmic antibodies(ANCA)associated vasculitis(AAV).Methods A total of 85 MPO-AAV patients treated in Zhumadian Central Hospital from June 2021 to December 2022 were selected to be included in the case group,and 30 healthy volunteers who were collected in hospital during the same period were included in the control group.The basic data of patients were collected,serum MPO was detected by ELISA,neutrophil membrane MPO and intracellular MPO expression were detected by flow cytometry.Peripheral blood neutrophil MPO expression and serum MPO level were analyzed in the two groups.Spearman correlation analysis was conducted to determine the relationship between peripheral blood neutrophil MPO expression and serum MPO level,and multiple regression analysis was performed to analyze the influencing factors of Birmingham vasculitis activity score-version 3(BVAS-V3).Results There was no statistical significance in MPO expression in peripheral blood neutrophils between the two groups(P>0.05).The MPO expression in peripheral blood neutrophils in the case group was lower than that in the control group(P<0.05).The serum MPO level of the case group was higher than that of the control group(P<0.05).There was negative correlation between MPO expression in neutrophils and serum MPO level(r=-0.711,P<0.05).Multiple regression analysis showed that BVAS-V3 was positively correlated with neutrophil MPO expression and MPO-ANCA expression(P<0.05),but was not significantly correlated with age,disease course,erythrocyte sedimentation rate,C-reactive protein,neutrophil count,neutrophil membrane MPO expression,and serum MPO expression(P>0.05).Conclusion In patients with MPO-AAV,intracellular MPO expression in peripheral blood of neutrophils is significantly increased,and positive MPO-ANCA may interfere with the expression of MPO in neutrophils,and MPO-ANCA expression in neutrophils is an independent factor affecting BVAS-V3.
