不同浓度TGF-β1对子宫内膜组织上皮细胞数量与转分化的影响研究

目的 探查不同浓度的转化生长因子-β1(transforming growth factor-β1,TGF-β1)对子宫内膜组织上皮细胞(endometrial epithelium cells,EECs)数量和转分化的影响。方法 收集河南省直属机关第二门诊部手术获取的正常子宫内膜组织40例,分离上皮细胞,根据随机数字表法,将细胞分为正常对照组(只加培养液)10份与实验组(加培养液和不同浓度的TGF-β1)30份,其中实验组又细分为三组,即低剂量TGF-β1组10份,中等剂量TGF-β1组10份,高剂量TGF-β1组10份,高、中、低剂量组分别给予TGF-β1(9 ng/mL、6 ng/mL、3 ng/mL)。测定活细胞数量、细胞悬液中 Ⅰ型胶原(Col-Ⅰ)与纤黏连蛋白(FN)含量以及细胞胞质中α-平滑肌肌动蛋白(α-SMA)和钙黏附蛋白-E(E-cadherin)的含量。结果 正常对照组FN分泌量为(0。469± 0。010)ng/L、Col Ⅰ分泌量为(0。532±0。033)ng/L;而各实验组测得的FN值与Col-Ⅰ值,差异有统计学意义(P<0。01);与正常对照组相比较,TGF-β1对E-cadherin及α-SMA的表达有相反的效果,且TGF-β1浓度不同,效果也不同,具体表现为:TGF-β1浓度逐步增加,E-cadherin在胞质中的量将逐渐减少,相反,α-SMA在胞质中的量将逐渐增多(P<0。05)。差异有统计学意义(F=145。125、179。511、P<0。01)。结论 不同浓度的TGF-β1能刺激子宫内膜组织上皮细胞数目增加,诱导其向肌成纤维细胞转变,且TGF-β1浓度越高,作用越强。
Effects of different concentrations of TGF-β1 on the number and transdifferentiation of epithelial cells in endometrial tissues
Objective To explore different concentrations of transforming growth factors-β1(transforming growth factor-β1.TGF-β1)the effect of on the number and transdifferentiation of endometrial epithelial cells(EECs)in en-dometrial tissue.Methods Totally 40 cases of normal endometrial tissue obtained from surgery in our hospital were col-lected,and epithelial cells were isolated.According to the random number table method,the cells were divided into 10 normal control groups(with only culture medium added)and 10 experimental groups(with culture medium and different concentrations of TGF)-β1)30 copies,of which the experimental group is further divided into three groups,namely low-dose TGF-β1 set of 10,medium dose TGF-β1 set of 10,high-dose TGF-β1 group of 10,with high,medi-um,and low dose groups receiving TGF-β1(9 ng/mL,6 ng/mL,3 ng/mL).Measure the number of living cells,the content of type Ⅰ collagen(Col Ⅰ)and fibronectin(FN)in cell suspension,and the content of cell cytoplasm α-Smooth muscle actin(α-SMA and E-cadherin).Results That the secretion of FN in the normal control group was(0.469±0.010)ng/L,and the secretion of Col Ⅰ was(0.532±0.033)ng/L;The difference between the FN values measured in each experimental group and the Col-Ⅰ values was statistically significant(P<0.01);Compared with the normal control group,TGF-β1 to E-cadherin and α-SMA expression has the opposite effect,and TGF-β1,differ-ent concentrations have different effects,specifically manifested as TGF-β1 As the concentration gradually increases,the amount of E-cadherin in the cytoplasm will gradually decrease.On the contrary,The amount of α-SMA in the cytoplasm will gradually increase(P<0.05).The difference was statistically significant(F=145.125,179.511,P<0.01).Conclusion Different concentrations of TGF-β1 can stimulate an increase in the number of epithelial cells in endometrial tissue,induce their transformation into myofibroblasts,and TGF-β1 the higher the concentration,the stronger the effect.
