目的 研究IL-17A和IL-17F基因三种常见的单核苷酸多态性(rs2275913G>A、rs3748067C>T和rs763780T>C)与胃癌发展之间的联系以及其与幽门螺杆菌感染的相互关系。方法 采用病例对照研究,选取2021年1月—2023年1月确诊的688例胃癌患者(试验组)和同一时期随机选取的688例健康体检者(对照组),抽取5 mL EDTA抗凝全血,按照制造商的说明用血液DNA提取仪提取受试者DNA,IL-17A rs2275913G>A,rs3748067C>T 和 IL-17Frs763780T>C 的基因分型用的是 Sequenom 公司的 GeneAmpPCR System 9700 和Dua1384孔板进行。结果 试验组和对照组在性别、年龄和吸烟史上差异无统计学意义(P>0。05);在直系亲属中,有患癌史,有幽门螺杆菌感染和饮酒史方面,差异有统计学意义(P<0。05)。试验组和对照组的白介素17A rs2275913G>A的基因型分布差异有统计学意义(P<0。05)。通过logistic回归分析,与GG基因型相比,携带IL-17rs2275913G>A GA与AA的个体与胃癌高风险相关,校正后的GA基因型OR(95%CI)值为1。48(1。03~2。06),校正后的AA基因型OR(95%CI)值为2。62(1。51~4。43)。本研究发现,幽门螺杆菌感染者中rs2275913的GA+AA基因型与胃癌高风险相关(OR=2。23,95%CI=1。29~3。79)。结论 在幽门螺杆菌感染者中,IL-17A rs2275913G>A的多态性与胃癌易感性显著相关。
Study on interleukin-17 gene polymorphism and susceptibility to gastric cancer
Objective To investigate the association between three common single nucleotide polymorphisms(SNPs)of IL-17A and IL-17F genes(rs2275913G>A,rs3748067C>T and rs763780T>C)and gastric cancer development,as well as their relationship with Helicobacter pylori infection.Methods A case-control study was conducted.688 pa-tients with gastric cancer diagnosed from January 2021 to January 2023(experimental group)and 688 healthy subjects randomly selected during the same period(control group)were selected.5 mL of EDTA anticoagulant whole blood was extracted with blood DNA extractor according to the manufacturers instructions.The genotyping of IL-17A rs2275913G>A,rs3748067C>T and IL-17Frs763780T>C was performed using the GeneAmp PCR System 9700 and Dual384 well plates of Sequenom.Results There were no statistically significant difference in gender,age and smoking history between the experimental group and the control group(P>0.05);there were statistically significant differences in the history of cancer,Helicobacter pylori infection and alcohol consumption in the immediate family(P<0.05).There was a statistically significant difference in the genotype distribution of IL-17A rs2275913G>A between the experimental group and the control group(P<0.05).Through logistic regression analysis,compared with GG genotype,individuals carrying IL-17 rs2275913G>A GA and AA were associated with a high risk of gastric cancer,with the adjusted OR(95%CI)value of GA genotype being 1.48(1.03-2.06),and the adjusted OR(95%CI)value of AA genotype being 2.62(1.51-4.43).In this study,the GA+AA genotype of rs2275913 was associated with a high risk of gastric cancer in patients with Helicobacter pylori infection(OR=2.23,95%CI=1.29-3.79).Conclusion IL-17A rs2275913G>A polymorphism is associated with gastric cancer susceptibility,especially in H.pylori infection.
