Relationship between miR-194 and miR-155 and hepatitis B virus DNA and prognosis in patients with viral hepatitis B cirrhosis
Objective To analyse the relationship between miR-194 and miR-155 and hepatitis B virus(HBV)DNA load in cirrhotic patients with viral hepatitis B,and to investigate the predictive value of miR-194,miR-155 and HBV DNA load in patients with poor prognosis.Methods Totally 200 patients with HBV infection who were not treated with antiviral therapy in the Second People's Hospital of Jiaozuo from June 2020 to June 2023 were selected as the subjects of investigation.According to the disease types,they were divided into HBV carrier group(n=59),chronic hepatitis B(CHB)group(n=77)and cirrhosis group(n=64).The patients with cirrhosis were followed up for 3 months and the prognosis was statistically analyzed.The expression levels of peripheral blood miR-194 and miR-155 and serum HBV DNA load were compared among the three groups of patients.Pearson correlation test was used to analyze the correlation between expression levels of miR-194 and miR-155 in peripheral blood and serum HBV DNA load in patients with cirrhosis.The expression levels of peripheral blood miR-194 and miR-155 and serum HBV DNA load in patients with cirrhosis with different prognosis status were compared.Receiver operating characteristic(ROC)curve was drawn and the area under the curve(AUC)was calculated to ana-lyze the predictive value of peripheral blood miR-194 and miR-155 and serum HBV DNA load on the poor prognosis of patients with cirrhosis.Results The expression levels of miR-194 and miR-155 in peripheral blood and serum HBV DNA load in cirrhosis group and CHB group were significantly higher than those in HBV carrier group(P<0.05),and the above indicators in cirrhosis group were significantly higher than those in CHB group(P<0.05).Pearson correlation test results showed that the expression levels of peripheral blood miR-194 and miR-155 were sig-nificantly positively correlated with serum HBV DNA load(P<0.05).The expression levels of peripheral blood miR-194 and miR-155 and serum HBV DNA load in poor prognosis group were significantly higher than those in good prog-nosis group(P<0.05).The AUCs of peripheral blood miR-194,miR-155,serum HBV DNA load and their combina-tion in predicting poor prognosis of patients with cirrhosis were 0.754,0.759,0.801 and 0.895 respectively.Conclusion The expression levels of miR-194 and miR-155 in peripheral blood of patients with viral hepatitis B cir-rhosis are significantly increased,and are closely related to the HBV DNA load and prognosis.miR-194 and miR-155 combined with HBV DNA load have certain predictive value on the poor prognosis of patients with viral hepatitis B cir-rhosis,and can provide reference for clinical practice.
