Correlation between gene promoter methylations of CALCA and DAPK and PAX1 and human papillomavirus E6/E7 mRNA detection and cervical lesions
Objective To detect the gene promoter methylations of calcitonin-related peptide alpha(CALCA),death-associated protein kinase(DAPK),paired box gene 1(PAX1)and human papilloma virus(HPV)E6/E7 mRNA,and to explore the correlation with cervical lesions.Methods Totally 192 patients with cervical lesions in the Puyang Oil-field General Hospital were selected from June 2021 to December 2023.All patients received colposcopy biopsy and de-tection of CALCA,DAPK,PAX1 gene promoter methylations and HPV E6/E7 mRNA.The detection of CALCA,DAPK and PAX1 gene promoter methylations and HPV E6/E7 mRNA were compared among different grades of cervical lesions.The correlation between gene promoter methylations of CALCA,DAPK and PAX1 and HPV E6/E7 mRNA detection and cervical lesions was analyzed.The diagnostic efficiency of CALCA,DAPK and PAX1 gene promoter methylations and HPV E6/E7 mRNA detection on high-grade squamous intraepithelial lesions(HSIL)and cervical cancer was evalua-ted.Results Among 192 patients with cervical lesions,there were 31 cases of cervical cancer,58 cases of HSIL,34 ca-ses of low-grade squamous intraepithelial lesions(LSIL)and 69 cases of chronic cervicitis.The positive rates of CAL-CA,DAPK and PAX1 gene promoter methylations and positive rate of HPV E6/E7 mRNA in cervicitis group,LSIL group,HSIL group and cervical cancer group were gradually increased(P<0.05).Spearman correlation analysis showed that the positive rates of gene promoter methylations of CALCA,DAPK and PAX1 and positive rate of HPV E6/E7 mR-NA were significantly positively correlated with cervical lesion grading(P<0.05).The AUC of combined detection for HSIL and cervical cancer was 0.806,which was higher than 0.781 of CALCA gene promoter methylation,0.726 of DAPK gene promoter methylation,0.735 of PAX1 gene promoter methylation and 0.722 of HPV E6/E7 mRNA detec-tion.Conclusion The more severe the cervical lesions,the higher positivity rates of CALCA,DAPK and PAX1 gene promoter methylations and HPV E6/E7 mRNA,and the combined detection of above indicators are helpful to enhance the diagnostic efficiency of high-grade cervical lesions and cervical cancer.
