Value of targeted high-throughput sequencing in diagnosis of pathogens of lung infection
Objective To investigate the application value of targeted next-generation sequencing technology in the di-agnosis of pulmonary infection pathogens,and to provide a theoretical basis for clinical diagnosis.Methods A total of 124 patients with suspected pulmonary infection who were admitted to Zhengzhou Yihe Hospital affiliated to Henan Uni-versity from January 2023 to April 2024 were enrolled and sent for tNGS and traditional testing(microbial culture+PCR).The distribution of pathogenic microorganisms was counted,the pathogenic detection results of the two methods were compared,the positive rate and consistency of tNGS and traditional detection were analyzed,and the factors affect-ing the positive detection rate of tNGS were analyzed.Results Among the 124 patients,tNGS detected 50 species and 371 strains of pathogens,among which bacteria had the highest detection efficiency(45.01%),and tNGS also detected some special pathogens.tNGS can improve the typing of both resistance genes and viruses.The positive detection rate of tNGS(91.94%)was higher than the traditional method(62.90%),and the difference was statistically significant(P<0.001).The detection rate of tNGS in bacterial,fungal,viral and co-infection was better than that of traditional detection,and the difference was statistically significant(P<0.05).Conclusion Compared with the traditional test,tNGS has a higher detection rate of pathogens in the samples of patients with suspected lung infection,and can be effec-tively applied to the etiological diagnosis of lung infection.
