两种提取三色堇DNA方法的比较

以三色堇叶片为材料,分别采用SDS方法和植物基因组DNA提取试剂盒法对三色堇叶片DNA进行提取,采用琼脂糖凝胶电泳法检测DNA的质量浓度和质量.结果表明:采用SDS方法提取的DNA质量浓度高,但纯度低,耗时长;试剂盒法获得的DNA纯度高,且该方法操作方便,耗时短,但费用较高.PCR扩增结果显示两种方法所提取的DNA均能满足基因扩增和分子标记检测的需要.
The comparison of DNA extraction methods of pansy
The leaves of pansy were used as materials, method of SDS and DNA secure plant kit were used to extract DNA from the pansy leaves. The concentration and quality of DNA were determined by agarose gel electrophoresis. The results showed that: The quality of DNA extracted by SDS method was high, but the purity was low and the time consuming was long. The high purity DNA could be obtained by DNA secure plant kit method, and the method was easy to operate and had short consumption time, but the cost was high. The results of PCR amplification showed that the DNA extracted from the two methods could meet the needs of gene amplification and molecular marker detection.
