Expression of S100A4 protein in conjunctival melanoma and its prognostic value
Objective To investigate the expression of S100A4 protein in conjunctival melanoma and its prognostic value.Methods Fifty-six patients with conjunctival melanoma who received surgical treatment in the Lingbao Fushoutang Ophthalmology Hospital and the First Affiliated Hospital of Zhengzhou University from January 2021 to January 2023 were selected as the study objects.Immunohistochemical SP method was used to detect the expression level of S100A4 protein in conjunctival melanoma tissue,according to the detection results,the enrolled patients were divided into S100A4 protein high expression group(n=35)and S100A4 protein low expression group(n=21),and the relationship between S100A4 expression level and overall survival time was compared.According to the prognosis of conjunctival melanoma patients,the enrolled patients were divided into good prognosis group(n=18)and poor prognosis group(n=38),and the risk factors that may affect the poor prognosis of conjunctival melanoma patients were analyzed,a Logistic regression prediction model was constructed according to risk factors,and receiver operating characteristic curve was used to evaluate the effectiveness of the prediction model.Results The median survival time and 1-year survival rate of patients with conjunctival melanoma were 10 months and 80.36%.The median survival time and 1-year survival rate(9 months,74.58%)of and patients with high expression of S100A4 protein in conjunctival melanoma tissue were lower than those of patients with low expression of S100A4 protein(10 months,90.47%;Log-rank χ2=3.485,P=0.001).The rates of low tumor differentiation,lymph node metastasis and high S100A4 protein expression in the poor prognosis group(68.42%,73.68%,71.05%)were higher than those in the good prognosis group(22.22%,27.78%,22.22%;χ2=10.481,P=0.001;χ2=10.635,P=0.001;χ2=11.785,P=0.001).Logistic multivariate regression analysis showed that tumor differentiation was low(OR=2.042,95%CI:1.350-2.734),lymph node metastasis(OR=1.876,95%CI:1.225-2.526)or S100A4 protein ex-pression was high(OR=1.828,95%CI:1.354-2.301)was a risk factor for prognosis in patients with conjunctival mela-noma.The sensitivity,accuracy,positive predictive value and area under the curve were 92.16%、90.02%、89.34%and 0.914,respectively,in predicting the prognosis of conjunctival melanoma patients.Conclusion Low tumor differentia-tion,lymph node metastasis and high expression of S100A4 protein are risk factors for poor prognosis of patients with con-junctival melanoma.The model constructed based on the above factors has good diagnostic performance in predicting poor prognosis of patients with conjunctival melanoma.
