目的:探讨加味茵陈蒿汤对湿热哮喘大鼠气道重塑的影响及其可能的机制.方法:50只健康雄性SD大鼠随机均分为正常组、湿热哮喘模型组、加味茵陈蒿汤中、高剂量组、地塞米松组(n=10).采用多因素干预建立湿热哮喘大鼠模型.HE染色观察大鼠肺组织形态,ELISA法测定各组大鼠血清白介素-6(IL-6)水平,免疫组织化学染色观察大鼠气道信号转导因子-3(STAT3)和胸腺活化调节趋化因子(TARC)表达水平.采用Image-Pro Plus 6.0测定支气管总管壁厚度(Wat/Pbm)及气道平滑肌厚度(Wam/Pbm).结果:模型组大鼠湿热哮喘表现明显,HE染色出现典型气道重塑改变.气道TARC表达和Wat/Pbm、Wam/Pbm与血清IL-6和气道STAT3表达呈正相关(P<0.05).与模型组比较,中药中剂量组大鼠气道IL-6/STAT3表达、气道TARC表达(P<0.05)和气道Wat/Pbm(P<0.05)、Wam/Pbm(P<0.05)值明显降低.结论:加味茵陈蒿汤中剂量早期干预能显著减轻湿热哮喘大鼠气道重塑,机制可能与抑制大鼠肺组织IL-6/STAT3通路的表达有关.
The Effect of Jiawei Yinchenhao Tang(加味茵陈蒿汤, JWYCHT) on Airway Remodelling in Rats with Acute Dampness-heat Asthma
Objective:To discuss the influence of JWYCHT on airway remodelling in rats with acute dampness-heat asthma and its possible mechanism.Methods:The 50 healthy male SD rats were randomly divided into control group,dampness-heat asthma model group and JWYCHT medium dose group,JWYCHT high dose group and dexamethasone group (n=10).The model rats of dampness-heat asthma were induced by multi-factors.The histological changes were evaluated by HE staining.The level of IL-6 in blood was measured by ELISA.The expression of STAT3 and TARC in lung tissue extracts was detected by immunohistochemistry technique.The airway total wall thickness (Wat/Pbm);the bronchial smooth muscle thickness (Wam/Pbm) were measured by using image analysis system.Results:The histological changes by HE staining showed the typical airway remodeling in dampness-heat asthma model group.The level of IL-6 and STAT3 showed positive correlations with TARC,Wat/Pbm and Wam/Pbm(P<0.05).JWYCHT medium dose group intervention in the early time of dampness-heat asthma could significantly reduce the expression of IL-6/STAT3 (P<0.05),airway TARC expression,Wat/Pbm and Wam/Pbm (P<0.05) values.Conclusion:JWYCHT can significantly reduce heat airway remodelling in dampness-heat asthma rats.Its possible mechanism may be related to inhibition of expression of IL-6/STAT3 pathway in pulmonary tissue of rats.
