Effect of Curcumin on Inhibiting Oral Submucosal Fibrosis by Regulating HIF-1α/miR-760/LTBP2 Axis
Objective:To explore the effects of curcumin on inhibiting oral submucosal fibrosis by regulating hypoxia-inducible factor-1α/micro RNA-760/laten transforming growth factor binding protein 2(HIF-1α/miR-760/LTBP2)axis.Methods:In the 40 SD rats,10 cases were randomly enrolled as normal group(without any treatment),and the other 30 cases were induced by arecoline to prepare the models of oral submucosal fibrosis.The 30 rats were randomly divided into model group,curcumin group and positive control group.Rats were treated with corresponding drugs respectively in curcumin group and positive control group.The rats in normal group and model group were administered intragastrically with the same volume of normal saline,once a day,continuously for 8 weeks.The degree of mouth opening,changes of buccal mucosa,expressions of fibrosis markers and HIF-1α/miR-760/LTBP2 signal axis in different groups were compared.Results:Compared with normal group,the degree of mouth opening was significantly decreased in model group(P<0.05),while score of buccal mucosa,expressions of TGF-β1,Col Ⅲ,IFN-γ,miR-760 mRNA,HIF-1 α mRNA,LTBP2 mRNA,HIF-1 αprotein,LTBP2 protein were increased in model group(P<0.05).Compared with model group,the degree of mouth opening was significantly increased in curcumin group and positive control group(P<0.05),while score of buccal mucosa,expressions of TGF-β1,Col Ⅲ,IFN-γ,miR-760 mRNA,HIF-1α mRNA,LTBP2 mRNA,HIF-1αprotein and LTBP protein were significantly decreased in curcumin group and positive control group(P<0.05).The curcumin group showed larger degree of mouth opening than positive control group(P<0.05),while lower score of buccal mucosa,expressions of TGF-β1,Col Ⅲ,IFN-γ,miR-760 mRNA,HIF-1α mRNA,LTBP2 mRNA,HIF-1α protein and LTBP2 protein than positive control group(P<0.05).Conclusion:Curcumin may inhibit oral submucosal fibrosis by decreasing expressions of HIF-1α,miR-760 and LTBP2.
