Effect and Mechanism of Jinshui Tingling Decoction(金水葶苓汤)on the Mouse Model of Pleural Effusion in Lung Cancer Transplant Tumors
Objective:To investigate the inhibitory effect of Jinshui Tingling decoction on pleural effusion by replicating the mouse model of LEWIS lung cancer transplant tumor pleural effusion.Methods:The pleural effusion lung cancer model of C57 mice was replicated by LEWIS cells chest injection.The effect of Jinshui Tingling decoction and Jinshui Tingling decoction combined with cisplatin injection on the volume of pleural effusion,number and quality of metastatic tumors in the chest wall metastases,and survival time was observed.Western blotting method was used to detect the expression levels of AQP1 protein in parietal pleural and VEGF protein in pleural effusion,to evaluate the therapeutic effect and mechanism of Jinshui Tingling decoction on pleural effusion mice with lung cancer transplant tumor.Results:Compared with the model group,the weight gain of mice was significantly increased in each drug delivery group,and the volume of pleural fluid,the number of metastases of the chest wall and the weight of metastases were significantly reduced in each drug delivery group.The drug delivery groups showed lower expression of VEGF and AQP1 protein than model group(P<0.05 or P<0.01),and Jinshui Tingling decoction+cisplatin injection group showed better effect than Jinshui Tingling decoction or cisplatin injection(P<0.05).Conclusion:Jinshui Tingling decoction can inhibit the generation of pleural effusion in malignant pleural effusion model mice,improve body weight,prolong survival time,reduce the number of chest wall tumor metastases,inhibit tumor growth,and decrease the expression levels of VEGF in pleural effusion and AQP1 protein in pleura.Moreover,Jinshui Tingling decoction combined with cisplatin injection shows better effect than Jinshui Tingling decoction and cisplatin injection alone.
