The Extraction Technology and Quality Standard of Scalding Drugs
Objective:To optimize the soaking extraction process of scalding drug and improve the quality standard of scalding drug.Methods:The concentration of impregnating solvent,impregnating time,impregnating temperature and the degree of comminution of raw materials were used as the factors,and the content of DiosgeninⅥ and alcohol-soluble extract were used as the evaluation indexes.The optimal soaking extraction process was optimized by L9(34)orthogonal test,and the Fangfeng(Saposhnikovia divaricate),Dahuang(rhubarb)and Jianghuang(turmeric)were qualitatively identified by Thin layer chromatography(TLC).The content of DioscoreaⅥ was determined by HPLC.Results:The optimum extraction process was to crush the decoction pieces of scalding medicine into fine degree of comminution and soak the powder with 50％ethanol as solvent at room temperature for 5 days.In the range of 19.307 15-154.457 20 μg/mL,there was a good linear relationship(r=0.999 9).The average recovery was 102.37％and RSD was 1.48％(n=6).The spots of Fangfeng(Saposhnikovia divaricate),Dahuang(rhubarb)and Jianghuang(turmeric)were clear in qualitative identification,and the separation effect was good.Conclusion:The optimized extraction process is stable and feasible,and the established quality control method shows good stability,reproducibility and specificity,which can be used as the basis for the quality control of scalding drugs.
