Exploring the Mechanism of Wenshen Xuanbi Decoction(温肾宣痹汤,WSXBD)in Treating Knee Osteoarthritis based on HPLC-Q-TOF-MS/MS and Network Pharmacology
Objective:To analyze the effective components of WSXBD using HPLC-Q-TOF-MS/MS,and explore the targets and pathways of WSXBD in treating knee osteoarthritis(KOA)using network pharmacology to investigate its mechanism of action.Methods:The chemical components of WSXBD were analyzed by liquid chromatography-quadrupole time-of-flight mass spectrometry(HPLC-Q-TOF-MS/MS).The active ingredients of WSXBD were retrieved from TCMSP V2.0 and screened for their corresponding targets.The targets related to KO A were obtained from databases such as"GeneCards".VennDiagram was used to obtain the overlapping targets of"traditional Chinese medicine compounds and diseases".The protein-protein interaction network was constructed using STRING database and Cytoscape software.The core target was selected,and gene ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed.Results:Using HPLC-Q-TOF-MS/MS analysis in positive and negative ion modes,a total of 24 compounds were identified in WSXBD,which were derived from Gouji(Cibotii Rhizoma),Fuzi(Aconiti Lateralis Radix Praeparata),Tianma(Gastrodiae Rhizoma),Shanyurou(Corni Fructus),Xixin(Asari Radix Et Rhizoma),Baizhu(Atractylodis Macrocephalae Rhizoma),Muxiang(Aucklandiae Radix),and Gancao(Glycyrrhizae Radix Et Rhizoma).A total of 181 active components and 134 corresponding protein targets of WSXBD were screened,and there were 72 core targets intersecting with 2 171 disease-related targets of osteoarthritis.The GO enrich-ment analysis obtained 76 biological process(BP)enrichment results,12 cell component(CC)enrichment results,and 29 molecular function(MF)enrichment results.The KEGG pathway enrichment analysis yielded a total of 27 related pathways.Conclusion:WSHXD can exert therapeutic effects on knee osteoarthritis through anti-inflammatory,immunomodulatory,anti angiogenic,and antioxidant effects.
