乳痛软坚片抗炎镇痛及调节免疫药效作用的实验研究

摘要

目的:探讨乳痛软坚片抗炎、镇痛及调节免疫的药效作用.方法:通过小鼠扭体实验、小鼠热板法实验,采用醋酸扭体及热板致痛模型,以扭体次数、痛域值为检测指标,观察乳痛软坚片的镇痛作用;通过大鼠肉芽肿实验、大鼠足肿胀实验,采用棉球植入诱导大鼠肉芽肿增生、角叉菜胶致炎模型,以肉芽重量、足跖肿胀度为检测指标,观察乳痛软坚片抗炎作用;通过小鼠血清溶血素实验、小鼠碳粒廓清实验,采用腹腔注射5%氯化钠溶液鸡红细胞混悬液、小鼠尾静脉注射20%印度墨汁(0.1 mL/10 g)制造小鼠免疫抑制模型,以血清溶血素、廓清指数(K)、吞噬指数(α)为检测指标,观察乳痛软坚片的调节免疫作用.结果:扭体实验中,乳痛软坚片高剂量组小鼠扭体潜伏期长于模型组,扭体次数少于模型组(P<0.01或P<0.05);乳痛软坚片中剂量组小鼠扭体次数少于模型组(P<0.05).热板法实验中,乳痛软坚片组高、中剂量组小鼠给药后1、2、3 h痛域值高于模型组(P<0.01或P<0.05).大鼠肉芽肿实验中,乳痛软坚片组高、中、低剂量组大鼠肉芽质量均低于模型组(P<0.01或P<0.05).大鼠足肿胀实验中,乳痛软坚片高剂量组大鼠在致炎后1、3、4 h足跖肿胀度低于模型组(P<0.05),乳痛软坚片中剂量组大鼠在致炎后3、4h足跖肿胀度低于模型组(P<0.05).小鼠血清溶血素实验中,乳痛软坚片高剂量组小鼠溶血素高于模型组(P<0.01).小鼠碳粒廓清实验中,乳痛软坚片高、中、低剂量组小鼠廓清指数、吞噬指数均高于模型组,差异均有统计学意义(P<0.01或P<0.05).结论:乳痛软坚片具有良好的抗炎、镇痛及调节免疫的药理作用.

Abstract

Objective:To explore the pharmacodynamic effects of anti-inflammatory,analgesic and im-munoregulatory effects of Rutong Ruanjians tablets.Methods:The analgesic effect of Rutong Ruanjian tablets was observed through mouse twisting experiment and mouse hot plate experiment,using acetic acid twisting and hot plate pain-causing model,with the number of times of twisting and the value of pain domains as the detection indexes.The anti-inflammatory effect of Rutong Ruanjian tablets was observed through rat granuloma experiment and foot swelling experiment,using the model of granuloma proliferation induced by implantation of cotton ball and inflammation caused by carrageenan gum,with the weight of granulomas and metatarsophalangeal swelling as the detection indexes.In the mouse serum hemolysin experiment and mouse carbon particle contouring experiment,the intraperitoneal injection of 5%sodium chloride solution chicken erythrocyte suspension,and mouse tail vein injection of 20%India ink(0.1 mL/10g)were used to create a mouse immunosuppression model,with serum hemolysin,contouring index(K)and phagocytosis index(α)as the test indicator,to observe the regulation of immunity of Rutong Ruanjian tablets effect.Results:In the torsion experiment,the high dose group of Rutong Ruanjian tablets showed longer torsion latency than model group,while less number of torsions than model group(P<0.01 or P<0.05);The medium dose group of Rutong Ruanjian tablets showed less number of torsions than model group(P<0.05).In the hot plate experiment,the high and medium dose groups of Rutong Ruanjian tablets showed higher 1 h,2 h and 3 h post-drug pain domain values than model group(P<0.01 or P<0.05).In the rat granuloma experiment,the high,medium and low dose groups of Rutong Ruanjian tablets group showed lower granuloma mass than model group(P<0.01 or P<0.05).In the foot swelling experiment of rats,the high dose group of Rutong Ruanjian tablets showed lower swelling degree of the foot and metatarsal than model group at 1,3 and 4 h after inflammation(P<0.05),and the middle dose group of Rutong Ruanjian tablets showed lower swelling degree of the foot and metatarsal than model group at 3 and 4 h after inflammation(P<0.05).In the serum hemolysin experiment,the high dose group of Rutong Ruanjian tablets showed higher hemolysin than model group(P<0.01).In the experiment of carbon granule contouring,the high,medium and low dose groups of Rutong Ruanjian tablets showed higher contouring index and phagocytic index than model group,and the differ-ences were statistically significant(P<0.01 or P<0.05).Conclusion Rutong Ruanjian tablets shows good pharmaco-logical effects of anti-inflammation,analgesia and immunomodulatory.
