Effect of Kaixuan Xiezhuo Recipe(开玄泄浊方)on Peripheral Blood Th Cell Balance and Related Cytokine Expression in Rats with Yang Deficiency Psoriasis
Objective:To investigate the effect of Kaixuan Xiezhuo recipe on T helper cell 17(Th17)/regulatory T cells(Treg)balance and expression of related cytokines in peripheral blood of Yang deficiency psoriasis rats.Methods:Totally 72 rats were randomly divided into blank control group(n=16)and model group(n=56).After all the rats'backs were depilated,rats were externally coated with Vaseline in blank control group,while the rats in model group were given 5%imiquimod cream externally as well as Dahuang(rhubarb)gavage to establish the SD rat model of Yang deficiency psoriasis.The 50 successfully modeled rats were randomly divided into model control group,Tripterygium wilfordii polyglycoside tablet group and Kaixuan Xiezhuo recipe low,medium and high dose groups,with 10 rats in each group.The corresponding drugs were given by gavage in every group,once a day for seven days.The psoriasis area and severity index(PASI)score on the back of rats were observed.The ratio of Th17/Treg in peripheral blood was detected by flow cytometry.IL-17 and TGF-β were detected by ELISA.The skin of the back was taken for pathological section.Results:The Tripterygium wilfordii polyglycoside tablet group and Kaixuan Xiezhuo recipe low,medium and high dose groups showed significantly lower PASI score,Th17/Treg ratio and IL-17 level than model control group(P<0.05),while higher level of TGF-β than model control group(P<0.05).Conclusion:The mechanism of Kaixuan Xiezhuo recipe in the treatment of Yang deficiency psoriasis may be related to the regulation of the proportion of Th17 and Treg cells and the expression of related factors.
