Radical-Scavenging Active in Kunbu(Laminariae Thallus)Based on Spectrum-Effect Relationship
Objective:To establish the spectrum-effect relationship of the radical-scavenging activity in Kunbu(Laminariae thallus),and screen out the components closely related to this effect,on the basis of the previous fingerprint study of Kunbu(Laminariae thallus).Methods:The radical-scavenging activities of 11 batches of Kunbu(Laminariae thallus)were determined by 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)radical.Based on previous UPLC fingerprint study,grey relation analysis(GRA)and partial least squares regression analysis(PLSR)were adopted to detect the spectrum-effect relationship of the radical-scavenging activity.Results:The 11 batches of Kunbu(Laminariae thallus)have an IC50 of 0.0392-0.1044 g/mL for the scavenging of DPPH radical,and 0.002 2-0.011 6 g/mL for scavenging of ABTS radical.Additionally,GRA showed that the characteristic peaks with a correlation degree greater than 0.8 for DPPH radical scavenging ability include X24,X21,X22,X4,X27,X28,X23,X29,X9,X18,X17,X12,X25,X20 and X10,and that the characteristic peaks with a correlation degree greater than 0.8 in the ability to clear ABTS free radicals include X18,X22,X9,X4,X24,X12,X17 and X27.PLSR showed that the characteristic peaks with a DPPH free radical scavenging ability VIP>1,include X28,X24,X27,X29,X30,X8 and X18,and that the characteristic peaks with ABTS free radical scavenging ability VIP>1,include X30,X25,X18,X24,X33 and X17.The common peaks were X18 and X24 in the above analysis results.Conclusion:The components represented by X18 and X24 may be the pharmacodynamic basis of the radical-scavenging activity of Kunbu(Laminariae thallus).
