Establishment of HPLC Fingerprints and Determination of Seven Constituents for Xinnaoxin Tablet(心脑欣片)
Objective:To establish HPLC fingerprint of Xinnaoxin Tablet,and to determine the contents of salidroside,tyrosol,gallic acid,ethyl gallic acid,p-coumaric acid,caffeic acid and chlorogenic acid.Methods:The 75%methanol extract was analyzed on WondaSil C18-WR column(4.6 mm×250 mm,5 μm),with mobile phase methanol(A)-0.1%phosphoric acid(B),gradient elution,volume flow 1.0mUmin and column temperature 35 ℃.The detection wavelength was 275 nm,with an injection volume of 20 μL.Cluster analysis and principal component analysis were performed for Xinnaoxin Tablet HPLC fingerprint.With gallic acid as internal standard,the contents of various components were determined by QAMS.Results:There were 24 common peaks in HPLC fingerprints of 11 batches of samples,with the similarity greater than 0.960.Each batch of samples was divided into three categories.The cumulative variance contribution rate of the four principal components was 95.045%,and the linear relationship of the seven components was good(r≥0.995).The average recoveries were 96.76%-104.02%,and RSD was 0.05%-1.48%.The result of QAMS is close to that of external standard method.Conclusion:The method is simple,reliable and reproducible,and it can be used for quality control of Xinnaoxin Tablet.
