叶下珠饮片质量标准优化研究

目的:优化叶下珠饮片的质量标准。方法:采用薄层色谱(TLC)法对叶下珠饮片进行定性鉴别;采用电子显微镜观察叶下珠饮片粉末及横切面特征;采用高效液相色谱法测定没食子酸、柯里拉京的含量;参照2020年版《中华人民共和国药典》测定叶下珠饮片中水分、总灰分、酸不溶灰分和醇溶性浸出物的含量。结果:TLC分析中,叶下珠饮片与对照药材在相近位置均显示紫红色斑点。显微鉴别中,饮片粉末、横切面所含各种组织结构均可作为鉴别依据。含量分析中,没食子酸的线性回归方程为Y=15 068X-14509(r=0。999 8),含量为0。0110%~0。258 2%;柯里拉京的线性回归方程为Y=38 265X-7 032(r=0。999 9),含量为0。126 7%~0。329 1%;水分、总灰分、酸不溶灰分及醇溶性浸出物含量分别为9。49%~11。29%、6。02%~9。92%、0。64%~1。67%、15。73%~30。25%。结论:该质量标准可为准确、全面控制叶下珠饮片质量提供技术支持。
0ptimization Research of Quality Standard of Yexiazhu(Phyllanthus urinaria L.)Decoction Pieces
Objective:To optimize the quality standard of Yexiazhu(Phyllanthus urinaria L.)decoction pieces.Methods:Thin layer chromatography(TLC)was used for qualitative identification for Yexiazhu(Phyllanthus urinaria L.)decoction pieces.Microscopy was used to observe the characteristics of the powder and cross-sectional tissue.High-performance liquid chromatography was used to determine the concentrations of gallic acid and corilatine.The concentrations of water,total ash,acid-insoluble ash and ethanol-soluble extractives were measured by the methods described in the 2020 edition of Chinese Pharmacopoeia.Results:The TLC qualitative identification showed that the fuchsia spots from the Yexiazhu(Phyllanthus urinaria L.)decoction pieces existed at the corresponding position compared with the controls.In the microscopic identification,all kinds of tissues and structures contained in the powder and cross section could be used as the identification basis.The results of quantitative analyses showed that the linear equation of gallic acid was Y=15 068X-14 509(r=0.999 8),and that the content was 0.011 0%-0.258 2%.Similarly,the linear equation of corilatine was Y=38265X-7 032(r=0.999 9),and the content was 0.126 7%-0.329 1%.The content of water,total ash,insoluble ash and ethanol-soluble extractives were 9.49%-11.29%,6.02%-9.92%,0.64%-1.67%and 15.73%-30.25%,respectively.Conclusion:This quality standard research could offer technical support for controlling the quality of Yexiazhu(Phyllanthus urinaria L.)decoction pieces accurately and comprehensively.
