Study on Quality Standard of Wuzhimaotao(Fici Hirtae Radix)Decoction Pieces
Objective:To improve the quality standard of Wuzhimaotao(Fici hirtae radix)decoction pieces(FRDP).Methods:Twenty-nine batches of FRDP were collected from different producing areas to perform appearance and property identification,microscopic identification and thin layer chromatography(TLC)identification.Moisture,total ash,acid-insoluble ash and water-soluble extract were detected,and the content of psoralen and bergapten were determined by HPLC.HPLC method was used to establish the characteristic chromatogram of 29 batches of FRDP.Using Psoralen(NO.4 peak)as reference,similarity evaluation was conducted by using Similarity Evaluation System for TCM Chromatographic Fingerprints(2012 edition),and the common peaks were determined.Results:FRDP was like round or irregular thick pieces,and the surface was gray brown to reddish-brown.Its phloem was easily shed from xylem.It was hard and brittle,cross-section fiber.Stone cells,calcium oxalate square crystals,starch granules,cork cells and so on could be seen in the power of FRDP.The results of TLC identification showed that there were spots of the same color in the chromatographic position corre-sponding to the chromatographic position of psoralen.The average contents of water,total ash,acid-insoluble ash,water soluble extract,psoralen and bergapten were 5.67%-1 1.29%,1.90%-3.85%,0.06%-0.69%,5.71%-19.17%,0.01%-0.36%,0.01%-0.18%,respectively.The average values were 8.02%,2.47%,0.23%,12.16%,0.17%,0.05%,respectively.It is preliminarily proposed that the content of moisture,total ash,and acid-insoluble ash should not exceed 12.0%,4.0%,0.7%,respectively.The water-soluble extract,psoralen,and bergapten should not be less than 6.0%,0.02%,0.01%,respectively.There were 11 common peaks in the characteristic chromatogram of FRDP,and the similarity was all over 0.900.The identified peaks of 4 and 5 were psoralen and bergapten.Conclusion:The established standard in this paper can provide reference for the scientific evaluation of the quality of FRDP.
