Determination of Total Polysaccharide Content of Inonotus Obliquus and Comparison of Content in Different Parts
Objective:To evaluate the quality of Inonotus obliquus,establish a method for the determination of total polysaccharide content in Inonotus obliquus,and compare the difference of total polysaccharide content in different parts of Inonotus obliquus.Methods:The content of total polysaccharides in Inonotus obliquus was determined by ultraviolet-visible spectrophotometry,and the content of total polysaccharides in the dark brown shell and the internal yellow contextus of Inonotus obliquus was compared.Results:The linear range of anhydrous glucose was 18.03~108.19 μg/mL,with a good linear relationship(r=0.999 1).The RSD of precision,repeatability and stability of total polysaccharide content was less than 3%(n=6),and the average recovery was 100.97%(RSD=2.86%).The total polysaccharide content of 23 batches of Inonotus obliquus was 24.71-90.33 mg/g.The total polysaccharide content of Inonotus obliquus in the dark brown shell was significantly higher than that in the inner contextus,and the difference was statistically significant(P<0.05).Conclusion:The established content determination method is simple,reproducible,stable and operable,which can provide reference for the quality control and quality standard improvement of Inonotus obliquus.
