基于hnRNPF调控的TLR4/NF-κB信号通路研究肾元颗粒对糖尿病肾脏病炎症的影响

目的:基于hnRNPF调控的TLR4/NF-κB信号通路探讨肾元颗粒对db/db糖尿病肾脏病小鼠炎症的影响及其作用机制。方法:将16只SPF级db/db小鼠随机分为模型组和肾元颗粒组,予0。2%腺嘌呤模型饲料;另取8只同背景系野生小鼠作为空白组,予普通饲料。肾元颗粒组予以肾元颗粒混悬液灌胃给药,模型组和空白组小鼠予等量双蒸水灌胃,连续12周。给药12周后代谢笼收集小鼠24 h尿液,取各组小鼠血清、肾脏组织。生化检测各组小鼠血清肌酐(SCr)、血尿素氮(BUN)、谷丙转氨酶(ALT);ELISA法检测尿液中性粒细胞明胶酶相关脂质运载蛋白(NGAL)水平;PAS染色光镜观察肾组织病理改变;R-T PCR法检测小鼠肾脏组织中TLR4 mRNA、IL-6 mRNA、IKKα mRNA、NF-κB P65 mRNA、TNF-α mRNA、hnRNPF mRNA表达水平;Western blotting检测肾脏组织中hnRNPF、TLR4蛋白表达及IKKα、NF-κB P65磷酸化水平。结果:模型组小鼠血清SCr、BUN、ALT显著高于空白组(P<0。05);肾元颗粒组小鼠血清SCr、BUN、ALT显著低于模型组(P<0。05)。模型组小鼠尿液NGAL水平显著高于空白组(P<0。05);肾元颗粒组小鼠尿液NGAL水平明显低于模型组(P<0。05)。与空白组比较,模型组小鼠肾脏组织病变严重;与模型组比较,肾元颗粒组小鼠肾脏病变有所改善。模型组小鼠肾脏组织中TLR4 mRNA、IL-6 mRNA、IKKα mRNA、NF-κB P65 mRNA、TNF-α mRNA相对表达量高于空白组(P<0。05),hnRNPF mRNA相对表达量低于空白组(P<0。05);肾元颗粒组小鼠肾脏组织中TLR4 mRNA、IL-6 mRNA、IKKα mRNA、NF-κB P65 mRNA、TNF-α mRNA相对表达量低于模型组(P<0。05),hnRNPFmRNA相对表达量高于模型组(P<0。05)。模型组小鼠肾脏组织中hnRNPF蛋白相对表达量低于空白组(P<0。05),TLR4蛋白相对表达量及IKKα、NF-κB P65磷酸化水平高于空白组(P<0。05);肾元颗粒组小鼠肾脏组织中hnRNPF蛋白相对表达量高于模型组(P<0。05),TLR4蛋白相对表达量及IKKα、NF-κB P65磷酸化水平低于模型组(P<0。05)。结论:肾元颗粒可能通过hnRNPF调控的TLR4/NF-κB信号通路,抑制db/db糖尿病肾脏病小鼠炎症反应,从而改善相关生化指标,减轻肾脏损害。
The Effect of Shenyuan Granules(肾元颗粒)on Diabetic Kidney Disease Inflammation Based on the hnRNPF-Mediated TLR4/NF-κB Signaling Pathway
Objective:Based on the hnRNPF-regulated TLR4/NF-κB signaling pathway,to explore the effect of Shenyuan granules on inflammation in db/db diabetic kidney disease mice and its underlying mechanisms.Methods:A total of 60 SPF-grade db/db mice were randomly assigned to model group and Shenyuan granules group,receiving 0.2%adenine model feed.Additionally,eight wild-type mice served as blank group and received regular feed.Mice were administered Shenyuan granules suspension by gavage in Shenyuan granules group,while an equivalent volume of double-distilled water by gavage in model group and blank group for 12 consecutive weeks.After 12 weeks of administration,metabolic cages were used to collect 24-hour urine from mice.After urine collection,serum and kidney tissues were obtained.Biochemical analysis included assessing serum creatinine(SCr),blood urea nitrogen(BUN)and alanine aminotransferase(ALT).ELISA was employed to measure neutrophil gelatinase-associated lipocalin(NGAL)levels in urine.Kidney tissue pathology was observed through PAS staining.Real-time PCR was used to detect expression levels of TLR4 mRNA,IL-6 mRNA,IKKαmRNA,NF-κB P65 mRNA,TNF-α mRNA and hnRNPF mRNA in mouse kidney tissues.Western blotting was performed to measure hnRNPF and TLR4 protein expression,as well as phosphorylation levels of IKKα and NF-κB P65.Results:The model group showed significantly higher levels of serum SCr,BUN and ALT than blank group(P<0.05).In contrast,The Shenyuan granules group showed significantly lower levels of SCr,BUN and ALT than model group(P<0.05).The model group showed significantly higher urinary NGAL level than blank group(P<0.05).The Shenyuan granules group showed significantly lower urinary NGAL level than model group(P<0.05).Compared to the blank group,mice in the model group showed severe kidney tissue damage,whereas the Shenyuan granules group showed some improvement compared to the model group.In terms of molecular markers,the model group showed higher relative expression levels of TLR4 mRNA,IL-6 mRNA,IKKαmRNA,NF-κB P65 mRNA and TNF-α mRNA than blank group(P<0.05),while lower hnRNPF mRNA expression than blank group(P<0.05).The Shenyuan granules group showed lower relative expression levels of TLR4 mRNA,IL-6 mRNA,IKKα mRNA,NF-κB P65 mRNA and TNF-α mRNA than model group(P<0.05),while higher hnRNPF mRNA expression than model group(P<0.05).The model group showed lower hnRNPF protein expression than blank group(P<0.05),while higher TLR4 protein expression and phosphorylation levels of IKKα and NF-κB P65 than blank group(P<0.05).The Shenyuan granules group showed higher hnRNPF protein expression than model group(P<0.05),while lower TLR4 protein expression and phosphorylation levels of IKKα and NF-κB P65 than model group(P<0.05).Conclusion:Shenyuan granules may potentially alleviate inflammation in db/db mice through the hnRNPF-mediated TLR4/NF-κB signaling pathway,improving relevant biochemical indicators and mitigating renal damage.
