Optimization of the Preparation of"Teng Medicine"(腾药)Cream by Response Surface Optimization Methodology and a Preliminary Study on Its Analgesic Effect
Objective:"Teng medicine"hot pack agent was produced as"Teng medicine"cream,and the analgesic effect of"Teng medicine"hot pack agent and"Teng medicine"cream were compared.It is a reference for the development and use of"Teng medicine".Methods:Optimum formulation of the"Teng medicine"cream base was determined by single-factor screening and Box-Behnken response surface methodology.The five aspects of the cream,including uniformity of appearance,centrifugal stability,spreadability,cold stability and heat stability,were used as evaluation indexes to comprehensively evaluate the prepared"Teng medicine"creams.The analgesic effects of"Teng medicine"hot pack agent and"Teng medicine"cream were compared and studied using the acetic acid torsion method,formaldehyde pain model and hot plate method.Results:The optimal prescription for"Teng medicine"cream is 0.29 g of drug,0.9 1g of stearic acid and 0.50 g of glycerylmonostearate.The pain inhibition rate of"Teng medicine"cream and"Teng medicine"hot pack agent was 28.77%and 28.74%respectively in the acetic acid torsion method.In the formaldehyde pain model,the average cumulative foot licking time for mice in phase Ⅰ and phase Ⅱ of"Teng medicine"cream was 28.83 s and 36.00 s respectively;The average cumulative foot licking time for mice in phase Ⅰ and phase Ⅱ of"Teng medicine"hot pack agent was 30.17 s and 38.25 s respectively.In the hot plate experiment,the final percentage increase in pain threshold was 90.54%for"Teng medicine"cream and 69.87%for"Teng medicine"hot pack agent.Conclusion:The process of preparing"Teng medicine"cream is feasible and its analgesic effect is comparable to that of"Teng medicine"hot pack agent.
