基于HPLC-Q-Exactive-MS/MS和GC-MS联用技术的蒙药通拉嘎-5化学成分研究

目的:系统分析蒙药通拉嘎-5化学成分,明确其化学物质组成。方法:采用HPLC-Q-Exactive-MS/MS和GC-MS联用技术对通拉嘎-5固有成分进行分析。结果:通过二级质谱结合CD软件和文献检索,共鉴定出105个化学成份,包括27个生物碱类,26个黄酮类,15个挥发油类,11个酚酸类,5个瑞诺烷类二萜类,5个有机酸类,3个多酚类,3个鞣质类,3个香豆素类和7个其他类。通过GC-MS法结合NIST数据库,共鉴定57个化合物,包括30个萜类、14个烃类、6个醛类、2个脂肪醇类、2个羧酸酯类、1个酮类、1个生物碱类和1个甾醇。结论:建立的HPLC-Q-Exactive-MS/MS 和GC-MS分析方法可对通拉嘎-5中化学成分进行快速识别和鉴定,可为系统阐明药效物质基础和质量控制提供实验参考。
Chemical Constituents of Tonglaga-5(通拉嗄-5)Based on HPLC-Q-Exactive-MS/MS and GC-MS
Objective:To analyze the chemical composition of Mongolian medicine Tonglaga-5 systematically in order to clarify its chemical composition.Methods:A comprehensive analysis of the intrinsic components of Tonglaga-5 was carried out by HPLC-Q-Exactive-MS/MS and GC-MS.Results:A total of 105 compounds were identified by secondary mass spectrometry combined with CD software and literature search,including 27 alkaloids,26 flavonoids,15 volatile oils,11 phenolic acids,5 ryanodine diterpenoids,5 organic acids,3 polyphenols,3 tannins,3 coumarins and 7 others.A total of 57 compounds,including 30 terpenoids,14 hydrocarbons,6 aldehydes,2 fatty alcohols,2 carboxylic acid esters,1 ketone,1 alkaloid and 1 sterol,were identified by GC-MS in combination with the NIST database.Conclusion:The HPLC-Q-Exactive MS/MS and GC-MS analytical methods established in this experiment can be used for the rapid identification and characterization of the chemical components in Tonglaga-5,and it can provide an experimental reference for the systematic elucidation of the pharmacophore basis and quality control.
