银屑病合并重度抑郁障碍差异表达基因及中药预测的生物信息学分析

目的:通过生物信息学方法筛选银屑病合并重度抑郁障碍的关键基因与信号通路,分析两者可能的共病机制,探究具有诊断价值的关键基因,并预测具有潜在疗效的中药。方法:从公共基因芯片数据库(GEO)中分别下载包含银屑病和重度抑郁障碍患者的基因数据集,分别应用Limma筛选出差异表达基因(DEGs),合并两种疾病的DEGs得到共同差异基因(co-DEGs),将交集基因导入metascape数据库进行GO和KEGG富集分析;基于STRING数据库和Cytoscape 3。9。0软件构建的蛋白相互作用网络筛选出关键基因,并使用OmicStudio在线工具对核心基因的诊断价值进行验证,最后通过Coremine Medical平台预测具有潜在疗效的中药。结果:共纳入1个银屑病数据集(GSE30999)、2个重度抑郁数据集(GSE19738和GSE76826)和2个验证数据集(GSE13355和GSE38206),得到1 554个银屑病DEGs和475个重度抑郁障碍DEGs,取交集后获得87个co-DEGs。GO和KEGG分析提示,co-DEGs主要富集在226个生物过程、18个细胞结构、34个分子功能及19条信号通路上。基于蛋白相互作用网络锁定了 9个可能的关键基因,经验证其中7个具有一定的诊断价值,并预测出46味具有潜在疗效的中药。结论:银屑病合并重度抑郁障碍的机制可能与NOD样受体信号通路及RIG-Ⅰ样受体信号通路相关,STAT1、DDX58、MX1、GBP1、IRF7、IFIT3、IFI44、RSAD2、ISG15可能是诊断和治疗的关键基因,生地黄、大青叶、黄芩、莪术、郁金、当归、人参等中药可能成为治疗的潜在药物。
Bioinformatics Analysis of Differentially Expressed Genes and Prediction of Traditional Chinese Medicine in Psoriasis Complicated with Major Depressive Disorders
Objective:To screen the key genes and pathways of psoriasis complicated with major depression disorders by bioinformatics method,to analyze the possible comorbidity mechanism,to explore the key genes with diagnostic value,and to predict the traditional Chinese medicine with potential effects.Methods:The gene data sets of patients with psoriasis and major depressive disorders were downloaded from Gene Expression Omnibus(GEO)database.Differentially expressed genes(DEGs)of those two diseases were screened by limma package,merge DEGs from the two diseases to obtain common differentially expressed genes(co-DEGs),and import the intersecting genes into the metascape database for GO and KEGG enrichment analysis.Key genes were selected based on the protein-protein interaction network constructed by STRING database and Cytoscape 3.9.0 software,and the diagnostic value of key genes was verified by OmicStudio online tool.Finally,the potential therapeutic traditional Chinese medicine was predicted by Coremine Medical platform.Results:A total of 1 psoriasis data set(GSE30999),2 major depressive disorders data sets(GSE19738 and GSE76826)and 2 validation data sets(GSE13355 and GSE38206)were included,resulting in 1554 psoriasis DEGs and 475 severe depression disorders DEGs,and 87 co-DEGs were obtained after intersection.GO and KEGG analysis indicated that co-DEGs were mainly concentrated in 226 biological processes,18 cell structures,34 molecular functions and 19 signaling pathways.9 possible key genes were identified based on protein-protein interaction network,7 among which proved to have diagnostic value,and 46 traditional Chinese medicine with potential curative effects were predicted.Conclusion:The mechanism of psoriasis complicated with major depression disorders may be related to the Nod-like receptor signaling pathway and the RIG-Ⅰ-like receptor signaling pathway.STAT1,DDX58,MX1,GBP1,IRF7,IFIT3,IFI44,RSAD2,and ISG15 may be the key genes in diagnosis and treatment.Traditional Chinese medicine such as Shengdihuang(Radix Rehmanniae),Daqingye(Isatidis Folium),Huangqin(Scutellariae Radix),Ezhu(Curcumae Rhizoma),Yujin(Curcumae Radix),Danggui(Angelicae Sinensis Radix),and Renshen(Ginseng Radix)may be potential drugs for treatment.
