摘要

目的:观察毛郁金提取物的抗炎作用,并探讨其作用机制.方法:采用巴豆油致小鼠耳肿胀实验、蛋清致小鼠足趾肿胀实验、冰醋酸致小鼠毛细血管通透性实验,观察毛郁金提取物的抗炎作用;通过脂多糖(LPS)诱导小鼠急性肾损伤(AKI)模型,采用酶联免疫吸附法(ELISA)检测AKI小鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)的含量,并检测肾组织TNF-α含量、肾脏系数.结果:毛郁金提取物高、中、低剂量组巴豆油致小鼠耳肿胀度低于空白对照组(P<0.05或P<0.01),蛋清致炎0.5、1.0、2.0 h后小鼠足趾肿胀度低于空白对照组(P<0.05或P<0.01),冰醋酸致小鼠毛细血管通透性低于空白对照组(P<0.05或P<0.01);毛郁金提取物高、中、低剂量组小鼠血清TNF-α、IL-6含量低于模型对照组(P<0.05或P<0.01),血清IL-10含量高于模型对照组(P<0.05或P<0.01),肾组织TNF-α含量及肾脏系数低于模型对照组(P<0.05或P<0.01).结论:毛郁金提取物具有明显的抗炎作用,其作用机制可能与降低TNF-α、IL-6含量,提高IL-10含量,调控炎症与抗炎之间的失衡有关.

Abstract

Objective:To study the anti-inflammatory effect and mechanism of Maoyujin(Curcuma aromatica Salisb)extract.Methods:The mice ear swelling by croton oil,mice toe swelling by egg white,and mice peritoneal capillary permeability by acetic acid were used to observe the anti-inflammatory effect of Maoyujin(Curcuma aromatica Salisb)extract.The acute kidney injury(AKI)of mice was induced by lipopolysaccharide(LPS).The levels of serum tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-10(IL-10)in AKI mice were detected by enzyme-linked immunosorbent assy(ELISA),and TNF-α level in renal tissue and renal coefficient were detected.Results:Compared with blank control group,the high-dose,middle-dose and low-dose of the Maoyujin(Curcuma aromatica Salisb)extract significantly reduced the mice ear swelling by croton oil(P<0.05 or P<0.01),reduced the mice toe swelling by egg white for 0.5,1.0,2.0 hours(P<0.05 or P<0.01),reduced the mice peritoneal capillary permeability by acetic acid(P<0.05 or P<0.01);Compared with model control group,the high-dose,middle-dose and low-dose of the Maoyujin(Curcuma aromatica Salisb)extract significantly decreased the levels of serum TNF-α,IL-6 in AKI mice(P<0.05 or P<0.01),increased the levels of serum IL-10(P<0.05 or P<0.01),decreased the levels of renal tissue TNF-α and renal coefficient(P<0.05 or P<0.01).Conclusion:The Maoyujin(Curcuma aromatica Salisb)extract has significantly anti-inflammatory effect.The mechanism may be involved in decreasing the levels of TNF-α,IL-6,increasing the levels of IL-10,and regulation the imbalance between inflammation and anti-inflammatory.
