Protective Effect of Acupuncture and Moxibustion on Articular Cartilage in Rats with Knee Osteoarthritis through Regulation of JAK2/STAT3 Pathway Based on Liver Theory and Treatment
Objective:To explore the protective effect of acupuncture and moxibustion on the articular cartilage of rats with knee osteoarthritis(OA)by regulating the JAK2/STAT3 pathway based on liver theory and treatment.Methods:Totally 60 SD rats were randomely divided into control group,OA group,OA+acupuncture group,20 rats in each group.The right knee OA model was surgically constructed.The rats in the OA+acupuncture group received acupuncture at acupoints selected from the liver theory for 4 weeks.Cartilage degeneration,joint inflammation,and JAK2/STAT3 transcription and protein levels were detected.Results:OA group showed higher OARSI score than control group(P<0.05).OA+acupuncture group showed lower OARSI score than OA group(P<0.05).OA group showed lower thickness of hyaline cartilage(HC)than control group,while higher thickness of calcified cartilage(CC)than control group(P<0.05).OA+acupuncture group showed higher thickness of HC than OA group,while lower thickness of CC than OA group(P<0.05).OA group showed higher IL-1β,IL-6,JAK2 mRNA,STAT3 mRNA,JAK2 protein and STAT3 protein than control group(P<0.05).OA+acupuncture group showed lower IL-1β,IL-6,JAK2 mRNA,STAT3 mRNA,JAK2 protein and STAT3 protein than OA group(P<0.05).Conclusion:Acupuncture and moxibustion based on acupoints selected from liver theory play a protective role on knee articular cartilage in knee OA rats by regulating JAK2/STAT3 pathway.
