基于超高效液相色谱-质谱联用技术鉴别蝉蜕及其制剂的真伪

目的:建立专属的鉴别蝉蜕真伪方法,对蝉蜕及其制剂进行质量评价。方法:采用超高效液相色谱(UPLC)-质谱(MS)串联法,以Shimadzu Scepter C18(2。1 mm×100。0 mm,1。9 μm)为色谱柱;以乙腈(A)-0。1%甲酸溶液(B)为流动相,梯度洗脱。采用三重四极杆质谱检测器,电喷雾正离子模式(ESI+),进行多反应监测(MRM),选择特征肽[质荷比(m/z):493。77]特征峰作为检测离子,对蝉蜕药材及其制剂进行测定。结果:30批蝉蜕药材、30批蝉蜕提取物、5批成方制剂均能检出特征肽(m/z:493。77)色谱峰。震旦马蝉、螗蝉属、僚蝉、蟪蛄、鸣鸣蝉均未检出相应的色谱峰。结论:该方法能快速准确鉴别蝉蜕药材及其制剂,可为蝉蜕质量评价和基础研究提供参考。
Authenticity Identification of Changtui(Cicadae Periostracum)Based on UPLC-MS
Objective:To establish an exclusive identification method of Changtui(Cicadae Periostracum)and evaluate its preparation quality.Methods:The UPLC-MS analysis was performed with Shimadzu Scepter C1s(2.1 mm×100.0mm,1.9 μm)column.The mobile phase was consisted of acetonitrile(A)and 0.1%formic acid(B)at a gradient mode.A triple quadrupole mass spectrometry was used with multiple reaction monitoring(MRM)and electrospray positive ion mode(ESI+).The characteristic peptide(m/z:493.77)was selected to detect Changtui(Cicadae Periostracum)and its preparation.Results:The characteristic peptide(m/z:493.77)was detected in 30 batches of Changtui(Cicadae Periostracum),30 batches of Changtui(Cicadae Periostracum)extractive and 5 batches of Changtui(Cicadae Periostracum)prescription preparation.The characteristic peptide(m/z:493.77)were not detected in Platylomiapieli Kato,Tanna Distant,Lyristes Horvath,Platypleurakaempfei(Fabricius)and Meimunamongolica(Distant).Conclusion:The method was used to rapidly and accurately identify the Changtui(Cicadae Periostracum)and its prescription preparation,providing reference for quality evaluation and basic research.
