Effects of Different Preparation Techniques on Cross-Sectional Tissues of Three Medicinal Plants
Objective:To select the most suitable slicing method for different medicinal parts of different medicinal plants[Paris polyphylla var.yunnanensis(Franch)Hand.-Mazz.,Schefflera elliptica(Blume)Harms,Dendrobium officinale Kimura & Migo.],then to improve the efficiency of cross-sectional tissue slicing preparation.Methods:Fresh materials were treated with different anti icing crystal protectants(untreated,sucrose,and glycerol)and frozen sectioning.Using glycerol to soften the dry material and conducting paraffin sectioning.Compare the production effects of different pre-processing conditions and two production methods.Results:Among the fresh materials,the use of glycerol solution as an ice crystal protectant is more effective for the stems and leaves of Schefflera elliptica(Blume)Harms,and the roots,stems,and leaves of Paris polyphylla var.yunna-nensis(Franch)Hand.-Mazz.,while sucrose phosphate buffer solution is suitable as a protectant for Dendrobium officinale Kimura & Migo.The fibrous roots of Paris polyphylla var.yunnanensis(Franch)Hand.-Mazz.can be sliced with paraffin to obtain intact tissue structures.In addition,by softening with glycerol solution,the dry rhizomes of Paris polyphylla var.yunnanensis(Franch)Hand.-Mazz.and the dry leaves of Schefflera elliptica(Blume)Harms.can obtain high-quality paraffin sections.Conclusion:Fresh medicinal herbs with slightly harder texture can achieve ideal results by treating them with glycerol solution and using frozen slices.Fresh medic-inal materials with soft texture can be frozen and sliced without any treatment or soaked in sucrose phosphate buffer.Dry medicinal materials or starch containing medicinal materials can achieve ideal slicing effects by using paraffin sectioning.
