Effects of Yinyanghuo(Epimedium),Dangui(Angelica Sinensis)and Their Combination Drugs on TGF-β1/Smads Signaling Pathway in Rats of Postmenopausal Osteoporosis
Objective:To investigate the effect of Yinyanghuo(Epimedium),Dangui(Angelica sinensis)and their combination on the prevention and treatment of postmenopausal osteoporosis(PMOP)through TGF-β1/Smads signaling pathway.Methods:A total of 42 female infertile rats were divided into sham operation group(n=7)and combined model group(n=35).Disease-associated postmenopausal osteoporosis rat model was established by bilateral ovariectomy combined with intramuscular injection of dexamethasone.After 6 weeks of modeling,30 combined model group rats were randomly divided into model group,Yinyanghuo(Epimedium)group(Epimedium group),Dangui(Angelica sinensis)group(Angelica sinensis group),Yinyanghuo(Epimedium)and Dangui(Angelica sinensis)combination group(combination group),and estradiol valerate group,6 rats in each group.Each group was given corresponding medication treatment for 8 weeks.At the end of treatment,the morphology of bone tissue was observed by HE staining.The serum level of TGF-[31 was detected by ELISA.The protein expressions of TGF-β1,Smad2,Smad3 and Smad7 were detected by immunofluorescence and the expressions of TGF-β1 mRNA,Smad2 mRNA,Smad 3 mRNA and Smad7 mRNA were detected by qRT-PCR.Result:The model group showed lower expression levels of TGF-β1 and Smad2/3 than sham operation group(P<0.01),while higher expression level of Smad7 than sham operation group(P<0.01).After intervention,the expression level of TGF-β1 in serum was significantly up-regulated by the combination of the two drugs(P<0.01).The protein expression levels of TGF-[31 and Smad2/3 and expression levels of Smad2/3 mRNA in the femoral tissue were significantly up-regulated in each administration group(P<0.01),while the protein expression level of Smad7 and expression level of Smad7 mRNA in the femoral tissue were significantly down-regulated in each administration group(P<0.05).Compared with single administration,the effect of combi-nation administration was more significant(P<0.01).Conclusion:Yinyanghuo(Epimedium),Dangui(Angelica sinensis)and their combination can improve bone metabolism and repair bone microstructure,and its mechanism may be through regulating TGF-β1/Smads signaling pathway,and improving the abnormal expression of related cytokines,so as to play a role in bone protection and prevention of osteoporosis.
