Effect of Acupuncture on the Expression of Inflammatory Factors in Obese Mice Induced by High Fat Diet
Objective:To observe the therapeutic effect of acupuncture on high-fat diet induced obese mice and its effect on changes in serum inflammatory factor protein and mRNA expression levels,and to explore the mechanism of acupuncture in reducing body mass in obese mice.Methods:Totally 12 C57BL/6J mice were randomly selected from 50 as the blank group,fed with a normal diet,and the remaining 38 mice were fed with a high-fat diet for 12 weeks.After successful modeling,24 mice were randomly divided into model group and acupuncture group.After 4 weeks of treatment,the differences in body weight and serum lipid levels of mice were compared among the 3 groups.The morphology of liver and epididymal fat in each group was observed by HE staining,and the expression of IL-1β,IL-6,TNF-α and MCP-1 protein were detected by Western blotting and the expression of IL-1 β mRNA,IL-6 mRNA,TNF-α mRNA and MCP-1 mRNA were detected by real-time fluorescence quantitative PCR.Results:Compared with the model group,the body weight and the content of total cholesterol,triglycerides and low density lipoprotein were significantly reduced in the acupuncture group.The liver lipid metabolism was significantly improved in acupuncture group,and the size of adipocytes was significantly reduced in acupuncture group.The level of inflammatory factor,including IL-1β,IL-6,TNF-α and MCP-1,as well as the expression of IL-1β mRNA,IL-6 mRNA,TNF-α mRNA and MCP-1 mRNA decreased significantly in acupuncture group.Conclusion:Acupuncture can significantly reduce the weight of obese mice,improve their pathological damage,and down-regulate the levels of TC,TG and LDL in serum,the expression of inflammatory factor(IL-1β,IL-6,TNF-α and MCP-1 protein),and expression of IL-1β mRNA,IL-6 mRNA,TNF-α mRNA and MCP-1 mRNA,to achieve the goal of treating obesity.
