Protective Effect of Huangqin Decoction(黄芩汤)on Renal Function in Mice with Unilateral Ureteral Obstruction
Objective:To study the protective effect and mechanism of Huangqin Decoction(HQD)on renal function in mice with unilateral ureteral obstruction(UUO).Methods:Totally 48 C57BL/6 mice were randomly divided into sham operation group,model group,HQD low dose group and HQD high dose group.The drug was given by gavage for 14 days from the first day after operation,while the mice in sham operation group and model group were given normal saline in the same way.On the last day,the serum of mice was collected to measure the renal function indexes and the expression of inflammatory factors.The renal tissues of mice were collected for histopathological observation and Western blotting detection.Results:The model group showed higher serum levels of SCr and BUN than sham operation group(P<0.01).The HQD low dose group and HQD high dose group showed lower serum levels of SCr and BUN than model group(P<0.05 or P<0.01).The model group showed hiher serum IL-1β,IL-6 and MCP-1 levels than sham operation group(P<0.01).The HQD low dose group and HQD high dose group showed lower serum levels of IL-1 β,IL-6 and MCP-1 than model group(P<0.01).The model group showed higher extracellular matrix(ECM)protein alpha SMA,Collagen Ⅰ and VIM relative expression in kidney tissue,than sham operation group(P<0.01).The HQD low dose group and HQD high dose group showed lower relative expressions of α-SMA,Collagen Ⅰ and VIM protein in kidney tissue,than model group(P<0.05 or P<0.01).The model group showed higher relative expressions of TGF-[31 and Smad4 protein in kidney tissue,as well as p-Smad2/Smad2,p-Smad3/Smad3,p-NF-κB p65/NF-κB p65,and p-STAT3/STAT3 than sham operation group(P<0.01),while lower Smad7 protein relative expression than sham operation group(P<0.01).The HQD low dose group and HQD high dose group showed lower relative expressions of TGF-β1 and Smad4 protein in kidney tissue,as well as p-Smad3/Smad3,p-NF-κB p65/NF-κB p65,p-STAT3/STAT3 than model group(P<0.05 or P<0.01),while higher relative expression of Smad7 protein than model group(P<0.01).The HQD high dose group showed lower p-Smad2/Smad2 than model group(P<0.01).Conclusion:HQD may adjust the TGF-[31/Smad,NF-κB and STAT3 pathway to reduce the reaction of inflammation and fibrosis in mice with UUO,thus reverse kidney injury.
