Optimization of Extraction Process of Mabei Qingfei Granules(麻贝清肺颗粒)by Box-Behnken Response Surface Methodology and Study on Its Pharmacodynamics
Objective:To optimize the extraction process of Mabei Qingfei granules,and evaluate the pharmacological effect on the treatment of acute bronchitis.Methods:The effects of water addition,extraction time and soaking time on the contents of ephedrine hydrochloride,nemangiferin,amygdalin,mangiferin,liquiritin,coptisine hydrochloride,berberine hydrochloride,palmatine hydrochloride,glycyrrhizic acid and dry paste ratio were screened by single factor test.On this basis,the Box-Behnken response surface method combined with information entropy theory was used to design the experiment and calculate the comprehensive score.According to analyzing the regression equation model,the factors affecting the extraction effect were investigated,and the extraction process of Mabei Qingfei granules was optimized.The pharmacological effects of Mapei Qingfei granules on cough,phlegm and inflammation were studied by using concentrated ammonia water to induce cough,ear swelling caused by xylene and tracheal phenol red.Results:The optimum extraction parameters were added 8.0 times water,soaked for 49.0 min and extracted for 1.8 h.Mabei Qingfei granules can significantly reduce the number of cough caused by concentrated ammonia water,reduce ear swelling and significantly increase the secretion of phenol red in mice.Conclusion:The optimized extraction process is stable and reliable.Mabei Qingfei granules shows obvious antitussive,antiphlegm and anti-inflammatory effects.
