Determination of Content of Quality Markers of Rhododendron Dauricum L.and Study on Its Optimal Harvesting Period
Objective:To establish a method for determining the content of quality marker based on network pharmacology research for Rhododendron dauricum L.,and evaluate different harvesting periods accordingly.Methods:Agilent Extend-C18 chromatographic column was used,with a flow phase of 0.2%phosphoric acid(A)-acetonitrile solution(B)for gradient elution.The flow rate was 1.0 mL/min,with column temperature of 35 ℃,and the detection wavelength was 360 nm(0-35 min)and 296 nm(35-60 min).The injection amount was 10 μL.Results:There was good linear relationship between peak area and the concentrations of Hyperoside,Quercetin,Kaempferol,8-norrhododendron and rhododendron respectively at 12.32-123.20,20.00-200.00,3.28-32.80,9.32-93.20,120.56-1 205.60 μg/mL.The average recovery rates were respectively from 99.70-103.96%(RSD=1.85%),99.13%-102.61%(RSD=1.71%),98.10%-102.86%(RSD=1.76%),98.29%-100.85%(RSD=0.90%)and 98.76%-100.62%(RSD=0.66%).July is the optimal harvesting period.Conclusions:A HPLC method for the determination of hyperoside,Quercetin,Kaempferol,8-demethylrhododendron and rhododendron was established simultaneously,which makes the quality evaluation of Rhododendron dauricum L more convenient and comprehensive.The best harvest time evaluated by this method is reasonable,providing experimental basis for the secondary processing and application of Rhododendron.
