Exploring the Interventional Mechanism of Xingpi Jieyu Formula(醒脾解郁方)on Abdominal Aortic Vascular Endothelium in Depressed Rats from the Perspective of Inflammation and Oxidative Stress
Objective:To explore the mechanism of vascular endothelial injury in depressed rats and the preventive and therapeutic effects of Xingpi Jieyu Formula from the perspectives of inflammatory factors and oxidative stress.Method:Totally 72 Wistar male rats were randomly divided into blank group,model group,low-dose group of traditional Chinese medicine(low-dose group),medium-dose group of traditional Chinese medicine(medium-dose group),high-dose group of traditional Chinese medicine(high-dose group),and western medicine group,with 12 rats in each group.Except for the blank group,the other 5 groups were subjected to 6 weeks of chronic unpredictable mild stress(CUMS),and the rats were orally administered while under stress.At the end of the 2nd and 6th week,6 rats were randomly selected from each group and euthanized after anesthesia.The endothelial cells of the abdominal aorta were taken,and interleukin-4(IL-4),interleukin-6(IL-6),interleukin-10(IL-10),tumor necrosis factor-α(TNF-α),glutathione peroxidase(GSH-Px),and endothelial growth factor(VEGF)were measured by enzyme-linked immunosorbent assay(ELISA).The content of nitric oxide synthase(eNOS)was measured by immunofluorescence method.Results:In the 2nd week,the model group showed higher levels of IL-6 and TNF-α in the endothelial cells of the abdominal aorta than blank group(P<0.05 or P<0.01);The low-dose group and western medicine group showed lower levels of IL-6 and IL-10 in the endothelial cells of the abdominal aorta than model group(P<0.01);The medium-dose group showed lower levels of IL-4,IL-6,and IL-10 in the endothelial cells of the abdominal aorta than model group(P<0.01 or P<0.05);The high-dose group showed lower levels of IL-4,IL-6,IL-10,and TNF-α in the endothelial cells of the abdominal aorta than model group(P<0.01 or P<0.05).In the 6th week,the model group showed higher levels of IL-4,IL-6,IL-10,TNF-α,and GSH-Px in the endothelial cells of the abdominal aorta than blank group(P<0.05 or P<0.01),while lower levels of VEGF and eNOS than blank group(P<0.01 or P<0.05);The low-dose group showed lower level of IL-6 in the endothelial cells of the abdominal aorta than model group(P<0.01);The medium-dose group showed lower levels of IL-4,IL-6,and IL-10 in the endothelial cells of the abdominal aorta than model group(P<0.01 or P<0.05),while higher level of VEGF than model group(P<0.05);The high-dose group showed lower levels of IL-4,IL-6,IL-10,and GSH-Px in the endothelial cells of the abdominal aorta than model group(P<0.01 or P<0.05),while higher levels of VEGF and eNOS than model group(P<0.01 or P<0.05);The Western medicine group showed lower levels of IL-4,IL-6 and IL-10 in the endothelial cells of the abdominal aorta than model grou(P<0.01 or P<0.05).Conclusion:Both Xingpi Jieyu Formula and Sertraline Hydrochloride can inhibit endothelial inflammation and oxidative stress response in the abdominal aorta of depressed rats.Xingpi Jieyu Formula shows an advantage in increasing the content of vascular endothelial factors.
