Polymorphism of Exon 6 of PLK2 Gene and its Correlation with Amino Acid and Fatty Acid Content in Muscle of Yanhuang Cattle
To investigate the single nucleotide polymorphism of PLK2 gene and its correlation with meat quality traits of Yanhuang cattle,96 Yanhuang cattle were selected as the research objects,and the PLK2 gene polymor-phism was detected by PCR sequencing.The linkage relationship between gene polymorphism and muscle amino acid and fatty acid content was analyzed.The results showed that there was a mutation site(Chr20:20,849,922C>T)in the sixth exon of PLK2 gene of Yanhuang cattle,and three genotypes were produced,CC being the domi-nant genotype.The homozygosity and effective allele numbers at this locus were 0.635 and 1.573,respectively.The polymorphism information content was 0.298,indicating moderate polymorphism.Linkage analysis showed that the valine content of CC genotype was significantly higher than that of CT genotype.The contents of palmitic acid and palmitoleic acid in CC and CT genotypes were significantly higher than those in TT genotype,and oleic acid content was significantly higher than that in TT genotype.The content of linoleic acid in TT genotype was signifi-cantly higher than that in CC and CT genotypes,and the content of stearic acid and arachidonic acid was signifi-cantly higher than that in CC and CT genotypes.It was preliminarily proved that PLK2 gene could be used as a po-tential molecular marker for the improvement of meat quality traits in Yanhuang cattle.
