摘要
目的 本研究通过采用饮食失节、劳倦伤脾双因素法建立脾虚证动物模型,观察玛咖对脾虚小鼠物质、能量代谢的影响,"以证测性"探讨玛咖偏温药性与健脾功效的相关性.方法 雄性昆明小鼠72只适应性喂养1周后随机分为正常组、模型组、人参对照组(0.75 g/kg)、玛咖高剂量组(1.5 g/kg)、中剂量组(0.75 g/kg)、低剂量组(0.25 g/kg),每组12只.给药14天,建立由饮食失节、劳倦伤脾造成的脾虚证小鼠模型,观察玛咖对脾虚小鼠体重、力竭游泳时间、肝糖原(liver glycogen,LG)、肌糖原(muscle glycogen,MG)、乳酸(lacticacid,LA)、游离脂肪酸(free fatty acid,FFA)、肌酐(creatinine,CREA)、尿素氮(urea,UREA)、乳酸脱氢酶(lactate dehydrogenase,LDH)、肌酸激酶(creatine kinase,CK)及环核苷酸系统cAMP/cGMP比值的影响.结果 玛咖能够增加脾虚小鼠体重,缓解脾虚小鼠体力疲劳增加力竭游泳时间(P <0.01),促进MG、LG储存(P <0.05、P <0.01、P<0.001),抑制代谢产物LA、FFA、CREA、UREA的生成(P <0.05、P <0.01、P <0.001),促进能量代谢酶LDH、CK活性(P <0.05、P <0.01),升高环核苷酸系统cAMP/cGMP比值(P <0.01、P <0.001).结论 玛咖能够促进脾虚小鼠物质、能量代谢,减少脾虚小鼠代谢产物的堆积,玛咖药性偏温,对脾虚小鼠具有健脾益气功效.
Abstract
Objective In this study, animal models of spleen deficiency syndrome were established by two-factor methods of eating disorders and fatigue.The effects of Maca on substance and energy metabolism in model mice were observed, so as to explore the relevance between warm property and strengthening spleen function through "measuring property by syndrome".Methods Seventy-two male Kunming mice were randomly divided into normal group, model group, ginseng control group (0.75 g/kg), Maca high dose group (1.5 g/kg), medium dose group (0.75 g/kg), and low dose group (0.25 g/kg).After 14 days of administration, a spleen deficiency model caused by eating disability, fatigue and spleen was established.The effects of Maca on weight, exhaustion swimming time, liver glycogen (LG), muscle glycogen (MG), lactic acid (LA), free fatty acids (FFA), creatinine (CREA), urea (UREA), lactate dehydrogenase (LDH), creatine kinase (CK) and the ratio of c AMP/c GMPin cyclic nucleotide system were observed.Results Maca can increase the body weight of mice with spleen deficiency syndrome, and increase the exhaustion swimming time to relieve fatigue (P< 0.01).Maca can promote the storage of MG and LG (P< 0.05, P< 0.01, P< 0.001), and inhibit the production of metabolites such as LA, FFA, CREA and UREA (P< 0.05, P< 0.01, P< 0.001).Maca can promote the activities of energy metabolism enzymes like LDH and CK (P< 0.05, P< 0.01), increase the ratio of cAMP/cGMP (P< 0.01, P< 0.001).Conclusion Maca can promote the substance and energy metabolism of mice with spleen deficiency syndrome and reduce the accumulation of metabolites.Maca's TCM properties are relatively warm and strengthen the spleen.
