Effects of acupuncture pretreatment on TRPV1 and CGRP protein expression in rats with abolute ethanol-induced acute gastric mucosal injury
Objective To investigate the possible mechanism of acupuncture pretreatment against anhydrous ethanol-induced acute gastric mucosal injury.Methods Fifty-two SD rats were divided into four groups:normal group,model group,pre-moxibustion group and pre-acupuncture group,each group has 13 mice.Acupuncture pretreatment of Zhongwan and Zusanli,20 min/acupoint,1 time/d,continuous 7 days,absolute ethanol gavage to prepare an acute gastric mucosal injury mode.To evaluate gastric mucosal injury(UI)index,HE staining was used to observe the pathological changes of gastric mucosal tissue,ELISA method was used to observe the concentration of TNF-α,IL-6,EGF,TFF1 in serum.Immunohisto-chemistry was used to analyze the expression levels of TRPV1 and CGRP proteins in rat gastric mucosal tissue.Results Compared with the normal group,the UI index score of the rats in the model group was increased,and the pathological manifestations such as hyperemia and erosion appeared on the surface of gastric mucosal tissue,indicating that the model was successfully prepared,the concentrations of TNF-αand IL-6 were increased,the concentration of EGF was decreased(P<0.05),the concentration of TFF1 was decreased(P>0.05,no statistically significant),and the expression levels of TRPV1 and CGRP proteins were increased(P<0.05).EGF levels were increased(P<0.05),TFF1 concentrations were increased(P>0.05,not statistically significant),and the expression levels of TRPV1 and CGRP proteins in the pre-moxibustion group and the pre-acupuncture group were decreased(P<0.05),compared with the pre-acupuncture group,the concentrations of TNF-α and IL-6,EGF levels were increased(P<0.05),and TFF1 concentrations were increased(P>0.05,not statistically significant),the levels of TRPV1 and CGRP protein expression were decreased(P<0.05).Conclusion Acupuncture pretreatment may achieve protective effect on rats with acute gastric mucosal injury by regulating the expression levels of TRPV1 and CGRP proteins.
