针灸预处理对无水乙醇致急性胃黏膜损伤大鼠香草酸亚型1、降钙素基因相关肽蛋白表达的影响

目的 探讨针灸预处理抗无水乙醇致急性胃黏膜损伤的可能机制.方法 将52只SD大鼠分为四组:正常组、模型组、预灸组和预针刺组,每组13只.针灸预处理中脘、足三里,20分钟/穴、1次/天,连续7天,无水乙醇灌胃制备急性胃黏膜损伤模型,评估胃黏膜损伤(ulcer index,UI)指数,苏木精—伊红(hematoxylin-eosin staining,HE)染色法观察胃黏膜组织的病理改变,酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)观察血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素 6(interleukin-6,IL-6)、表皮生长因子(epidermal growth factor,EGF)、三叶因子1(trefoil factor 1,TFF1)的浓度,免疫组织化学法分析大鼠胃黏膜组织香草酸亚型 1(transient receptor potential vanilloid 1,TRPV1)、降钙素基因相关肽(calcitonin gene related peptide,CGRP)蛋白表达水平.结果 与正常组相比,模型组大鼠UI指数评分升高,胃黏膜组织表面出现充血、糜烂等病理表现,提示模型制备成功.与正常组相比,模型组大鼠TNF-α、IL-6浓度上升(P<0.05);EGF浓度下降(P<0.05);TFF1浓度有下降趋势,但无统计学意义(P>0.05);TRPV1、CGRP蛋白表达水平上升(P<0.05).与模型组相比,预灸组和预针刺组大鼠TNF-α、IL-6浓度下降(P<0.05);EGF水平上升(P<0.05);TFF1浓度有上升趋势,但无统计学意义(P>0.05);预灸组和预针刺组大鼠TRPV1、CGRP蛋白表达水平下降(P<0.05).与预针刺组相比,预灸组大鼠TNF-α、IL-6浓度下降(P<0.05);EGF水平上升(P<0.05);TFF1浓度有上升趋势,但无统计学意义(P>0.05);TRPV1、CGRP蛋白表达水平下降(P<0.05).结论 针灸预处理可能是通过调节TRPV1、CGRP蛋白表达水平实现对急性胃黏膜损伤大鼠保护效应的.
Effects of acupuncture pretreatment on TRPV1 and CGRP protein expression in rats with abolute ethanol-induced acute gastric mucosal injury
Objective To investigate the possible mechanism of acupuncture pretreatment against anhydrous ethanol-induced acute gastric mucosal injury.Methods Fifty-two SD rats were divided into four groups:normal group,model group,pre-moxibustion group and pre-acupuncture group,each group has 13 mice.Acupuncture pretreatment of Zhongwan and Zusanli,20 min/acupoint,1 time/d,continuous 7 days,absolute ethanol gavage to prepare an acute gastric mucosal injury mode.To evaluate gastric mucosal injury(UI)index,HE staining was used to observe the pathological changes of gastric mucosal tissue,ELISA method was used to observe the concentration of TNF-α,IL-6,EGF,TFF1 in serum.Immunohisto-chemistry was used to analyze the expression levels of TRPV1 and CGRP proteins in rat gastric mucosal tissue.Results Compared with the normal group,the UI index score of the rats in the model group was increased,and the pathological manifestations such as hyperemia and erosion appeared on the surface of gastric mucosal tissue,indicating that the model was successfully prepared,the concentrations of TNF-αand IL-6 were increased,the concentration of EGF was decreased(P<0.05),the concentration of TFF1 was decreased(P>0.05,no statistically significant),and the expression levels of TRPV1 and CGRP proteins were increased(P<0.05).EGF levels were increased(P<0.05),TFF1 concentrations were increased(P>0.05,not statistically significant),and the expression levels of TRPV1 and CGRP proteins in the pre-moxibustion group and the pre-acupuncture group were decreased(P<0.05),compared with the pre-acupuncture group,the concentrations of TNF-α and IL-6,EGF levels were increased(P<0.05),and TFF1 concentrations were increased(P>0.05,not statistically significant),the levels of TRPV1 and CGRP protein expression were decreased(P<0.05).Conclusion Acupuncture pretreatment may achieve protective effect on rats with acute gastric mucosal injury by regulating the expression levels of TRPV1 and CGRP proteins.
